Using a Compound Light Microscope

Purpose: The purpose of this lab is to 1. Learn the parts of a compound light microscope. 2. The functions of those parts. 3. Proper use and care of the microscope. 4. Learn the technique of preparing wet-mount slides.
Materials:
* Compound light microscope * Soft cloth * Microscope slide * Cover slip * Dropper * Scissors * Newspaper
Procedures:
Part A. Care of the Compound Light Microscope.
Step 1: Always carry the microscope with both hands.
Step 2: Grasp the arm of the microscope with one hand and place your other hand under the base.
Step 3: Always hold the microscope in a upright position so that the eyepiece cannot fall out.
Step 4: Place a microscope on your worktable or desk at least 10cm from the edge.
Step 5: Position the microscope with the arm facing you.
Step 6: Record the magnifications of your microscope in the Data Table.
Step 7: The total magnification of a microscope is calculated by multiplying the magnification of the objective by magnification of the eyepiece.
Part B. Use of a Compound Light Microscope.
Step 1: Look at the microscope from side. Locate the coarse adjustment knob that moves the objectives up and down. Practice moving the coarse adjustment knob to see how it moves the objectives with each turn.
Step 2: Turn the coarse adjustment so that the low-power objective is positioned about 3cm from the stage.
Step 3: Locate the revolving nosepiece.
Step 4: Turn the nosepiece until you hear the high-power objective click into position.
Step 5: When an objective clicks into position, it is in the proper alignment for light to pass from the light source through the objective into the viewer’s eye.
Step 6: Now turn the nosepiece until the low-power objective clicks back into position.
Step 7: Adjust the diaphragm to permit sufficient light to enter the microscope.
