    The Smiths do their groceries every weekend. These are elaborately planned with Mrs. Smith making a list of requirements for the entire family over the week. She never forgets to replenish their stock of Caribbean coffee as her husband, a writer, generally feels groggy and sleepy after lunch in the afternoon. The coffee keeps him awake at such times.     In this lab experiment we did several test to determine what our unknown bacteria was. To determine this we recorded the results of how the bacteria reacted to different media. Depending on the results of each test we could narrow down the different bacteria to determine what our unknown is. This experiment will also determine if our bacteria is a fermenter of sugars and if it is catalase positive. If the bacteria is a fermenter they will use the sugars to make ATP. If the bacteria is a fermenter of lactose/sucrose the EMB plate we used will “clearly differentiate between the colonies of lactose fermenting and non-fermenting microbes. In the same medium sucrose was also included to differentiate between coliforms that were able to ferment sucrose more rapidly than those that were unable to ferment sucrose” (Cheeptham & Lal, 2007).…

    Since microorganisms are not visible to the eye, the essential tool in microbiology is the microscope. One of the first to use a microscope to observe microorganisms was Robert Hooke, the English biologist who observed algae and fungi in the 1660s. In the 1670s, “Anton van Leeuwenhoek, a Dutch merchant, constructed a number of simple microscopes and observed details of numerous forms of protozoa, fungi, and bacteria” (Introduction to Microscopes, n.d.). During the 1700s, microscopes were used to further explore on the microbial world, and by the late 1800s, the light microscope had been developed. “The electron microscope was developed in the 1940s, thus making the viruses and the smallest bacteria (for example, Rickettsiae and Chlamydia) visible” (Introduction to Microscopes, n.d.). The studies of microorganisms were now possible thanks to these two powerful tools the light and electron microscopes. The time from the development of the light microscope to the electron microscope was more than a century apart thus giving us our two first branches of microscopes and a new way to explore what the unaided eye could not.…
