Discussion for the research paradigm
Input:
Having the information that collagen was abundant protein in vertebrates people are used to extract collagen from mammals. But due to negative effects of collagen from mammals, fish skin serves as another option because it also contains collagen. It is much safer for it’s being said that marine collagen are not associated with the risk of bovine spongiform encephalopathy (BSE) outbreaks which is common in pig, cow or any mammals’ skins and bones.
In extraction of collagen we can be able to produce two kinds of collagen where they only differ in solvent used in the process. Acid soluble collagen was extracted with acetic acid while pepsin soluble collagens are dissolved in pepsin. The researchers are to use apparatuses like freezer on the study because a cold environment is needed for the storage of the skin to maintain its properties. Another are centrifuge and pH meter. For testing purposes the use of Amino acid analyser, SDS–polyacrylamide gel electrophoresis, and differential scanning calorimeter (DSC) is to be followed.
Process:
It is indicated in the diagram on how we can be able to extract our collagen from Janitor fish skins and what tests are needed to determine the properties of the extracted Acid soluble collagen and Pepsin soluble collagen. To compare amino acid profiles similarities w/ a collagen we used Amino acid analysis by AOAC method. Then to determine peptide mapping we used SDS–polyacrylamide gel electrophoresis (SDS–PAGE). And lastly to know the denaturation temperature, and compare if it is close to the denaturation temperature of a collagen we used differential scanning calorimeter (DSC).
OUTPUT:
Upon accomplishing the process we are expected to have a product of acid soluble collagen and pepsin soluble collagen. It is then be evaluated and determine its properties for the comparison from its standard collagen content so therefore we can

The gelatin eventually solidifies even though Bromelian was present. This reaction occurs because Bromelian is in a cold environment which can affect the effect of the enzyme. If the tube was taken out of the cold environment Bromelian would continue breaking down the protein. The pH of HCL and fresh pineapple juice was 0.     From what we observe in the gel results there are four bands and one smear, in seven lines which every line present a different serum (except 6 +7 are same 1 +2). Each band and the smear present different compound of serum protein. The first thick band is albumin which is the major fraction in a normal SPEP, and according to its concentration in serum it has the thicker band as its accounts for about 60 percent of the protein found in serum.
