Abstract-

In this experiment we tested to see how well halobacterium grows in different levels of salinity. We found out that the higher salinity in the growth medium the better the halobacterium grows.

Background Information-

Halobacterium is a bacteria that is found in the great salt lake. Halobacterium is an extremeophile which means it thrives in extreme environments. In this case the extreme environment is an extremely salty environment.

Question-

How does salinity affect the growth of halobacterium?

Hypothesis-

I think that the higher the salinity of the growths medium the better the halobacterium will grow. I think this because halobacterium thrives in extremely salty environments.

Materials-

Complete Medium (CM)
Live halobacterium culture
3 Falcon tubes
Spectrophotometer
3 Cuvettes 1 ML Beral pipet Micropipet w/tips

Variables-

Manipulated -Salinity of growth medium

Responding

-Growth of halobacterium

Controlled

-Temperature of halobacteruim samples -Amount of halobacterium -Amount of growth medium

Day One Procedure-

Note: Each group will be assigned ONE concentration of CM (2.3 M, 2.8M, 3.3 M, 3.8 M, 4.3 M) and each group will prepare three test samples with halobacteria and one sample without Halobacteria as a control.

1. Fill the necessary information on the pre-made, color coded labels for all three Falcon tubes.
2. Add mL o your assigned concentration of CM into each of the four Falcon tubes using a Beral pipet five times, for a TOTAL of 5 mL.
When your group is called to the mictopipet station, add 100.0 mL of Halobacterium into three of the Falcon tubes using a micropipet. Do NOT touch the tips. Do NOT put Halobacterium in your control Falcon tube.
3. Dispose of the micropipet tips in the disposal cup at he mictopipet station after putting together all three
