Problem: Before the lab, one should understand:
• The general functions and activities of enzymes;
• The relationship between the structure and function of enzymes
• The concept of initial reaction rates of enzymes;
• How the concept of free energy relates to enzyme activity;
• That change in temperature, pH, enzyme concentration, and substrate concentration can affect the initial reaction rates of enzyme-catalyzed reactions; and
• Catalyst, catalysis, and catalase

After the lab, one should be able to:
• Measure the effects of changes in temperature, pH, enzyme concentration, and substrate concentration on reaction rates of an enzyme-catalyzed reaction in a controlled experiment; and
• Explain how environmental factors affect the rate of enzyme-catalyzed reactions.

To establish a baseline, start by putting 10 mL of 1.5% H2O2 into a clean glass beaker. Add 1 mL of H2O (instead of enzyme solution). Add 10 mL of H2 SO4 (1.0 M). Mix well. Remove a 5-mL sample. Place this 5-mL sample into another beaker and assay for the amount of H2O2 as follows. Use a syringe to add KMnO4 , a drop at a time, to the solution until a persistent pink or brown color is obtained. Remember to gently swirl in the solution after adding each drop. Check to be sure that you understand the calibrations on the syringe. Record your reading.

    In regards to peroxidase, several points can be made about the overall effects of these factors. Factors such as temperature and pH have been shown to have an effect on the performance of enzymes (Vishwanatha, K.S., Food Chemistry. 2: 402-407; 2009). Adjusting the temperature will produce one of two results; if the enzymatic reaction occurs and the enzyme is colder than the optimum temperature, fewer collisions between enzyme and substrate will occur thus reducing the rate of the reaction. On the other hand, if the enzyme has been heated to a higher temperature than the optimum but not high enough to denature the enzyme, more of these collisions will occur and the reaction rate should speed up (Table 2, Graph 2). The second of the common factors is pH. Everything in the world has extremes on each end of the spectrum where the body, cell, etc. stops reacting to its environment. This goes for enzymes as well. If a solution is too basic or too acidic in comparison to the enzymes optimum pH, the enzyme will begin to degrade until it eventually stops reacting all together. It is important that students and biologists understand these and other optimal conditions for enzymes to react in.     How does changing the surroundings of enzymes affect their reaction rate? The purpose of the experiment is to determine how different abiotic conditions affect the rate at which enzymes accelerate/cause reactions In this lab students measured the height of the foam after catalysis between catalase (enzyme) and 7 other (solutions) to determine which solution had the fastest reaction rate.. The control variable of the experiment would be the solution of only hydrogen peroxide, water, and catalase. If I change the environment of the catalyst by adding room temperature , boiling , and freezing cold Solutions into three individual test tubes and measured the height of the foam then freezing cold Solution would have the highest rate of reaction this is because the higher the temperature the weaker the hydrogen bond.
