HYDROLYSIS AND ANALYSIS OF CARBOHYDRATES

Methodology

Materials:
|1% solutions: | conc. H2SO4 |
|glucose, fructose, maltose, sucrose, lactose, |Molisch reagent |
|agar-agar, gum arabic,glycogen, cotton, |I2 in KI solution (Lugol’s iodine reagent) |
|starch |Benedict’s reagent |
|10% HOAc solution, |Barfoed’s reagent |
|conc. HCl |Orcinol reagent |
|solid Na2CO3 |conc. HNO3 |
|dilute NaOH | |

Equipment: hot plate

Procedure:
A. Hydrolysis 1. For 1% sucrose, lactose and maltose solutions a. Measure out 5 ml 1% solution in a small beaker. b. Add 3 drops conc. HCl and boil for 5 minutes. c. Cool & neutralize with dilute NaOH. Set aside for part B. 2. For 1% starch, agar-agar, gum arabic, glycogen, cotton and starch a. Measure out 5 ml 1% solution in a small beaker. b. Add 5 drops conc. HCl. c. Cover the mouth of the test tube with a marble and boil in a water bath for 30 minutes. d. Neutralize with solid Na2CO3. Set aside for part B. 3. For polysaccharide extract a. Repeat
