Tamarindus Indica Case Study
Objectives of the Study
This study evaluates the genotoxicity, and teratogenicity/antiteratogenicity of the methanol leaf extracts of Tamarindus indica (TE). In particular, the study aims to: determine the genotoxicity of TE in terms of: mitotic index chromosomal aberrations micronuclei formation assess the developmental toxicity and teratogenicity potential of TE to zebrafish embryos; and verify the potential of TE to rescue zebrafish embryos from ethanol-induced teratogenicity.

Place and Time of Study:
The genotoxicity and teratogenicity/antiteratogenicity studies will be conducted in Caranglaan District, Dagupan City, and Don Mariano Marcos Memorial State University (DMMMSU) SLUC in Agoo, La Union, and DMMMSU MLUC in San Fernando,

84 bulbs of the aggregatum variety, locally known as sibuyas Tagalog, also known as Allium ascalonicum (Matias & Fontanilla, 2011), will be bought from the local market in Dagupan City. Dried scales will be removed and the bottom plate scraped, leaving the ring of root primordia intact, to stimulate root growth. Onions will be suspended to allow germination in 220-mL glass jars half-filled with mineral water for 72h in a 12h light/12h dark cycle at room temperature. For each series, the onion with the poorest root growth will be discarded.
The onions will then be transferred to glass jars containing 0.625, 1.25, 2.5, 5, and 10 mg/mL of T. indica methanol leaf extracts to which the roots will be exposed for 48 h in the same light-dark cycle. After the exposure of the bulbs to the different treatments, root lengths will be measured with a ruler, and will be used as an index of general toxicity. For each bulb, three slides will be made from three root tips 1-3 mm long. Root tips will be stained with a few drops of 2% acetocarmine for 30 min, and passed over the flame for 10 seconds (Crispino, 2016). Excess stain will be removed with tissue paper. A cover slip will be placed on the slide, which will then be hit with a bouncing motion of the rubber end of a pencil to squash the root tips.
