Enzyme Activity Lab Report
Temperature and Enzyme Activity

Aim: To investigate the effect of temperature on enzyme activity

Hypothesis: As the temperature deviates from 40°C the activity will lower

Equipment: – Chemicals: – Milk – Junket tablets – Hot water – Ice – Test tubes – Stopwatch – Measuring cylinder

Risk Assessment:
|Hazard |Risk |Prevention |
|Hot Water |Hot water splashing or spilling on |Safety glasses and aprons were worn |
| |skin | |
|Glassware

Junket tablet was crushed in a pestle and mortar and measured into 5gram portions. 2. Prepared beakers with water at temperatures 20, 30, 40, 50 and 60 degrees Celsius using Crushed ice to sustain the lower temperatures and adding hot water to reach higher temperatures. 3. 10mL of milk was poured into the test tubes using the measuring cylinder for accurate amounts then label the test tubes to indicate what temperature they will be subjected to and if they are to receive the enzyme. 4. Place 2 test tubes in each of the Beakers – one to receive the enzyme and one as a control. A thermometer was placed in each test tube. 5. The Junket was added and the timer started, tipping the test tube once every test seconds to monitor progress. 6. The timer was stopped when the Milk coagulated and the time was …show more content…

Purpose/Problem: There are four parts to the Enzyme Catalyst lab - Activity A, B, C, and D. In activity A, the characteristics of enzyme actions will be observed. The main purposes are to determine the rate of an enzyme catalyzed reaction, to study the characteristics of an enzyme mediated reaction, and to observe the effect of heat on enzyme activity. The purpose of activity B is to use the Titration Protocol to determine the initial amount of H2O2 present in a solution. The amount will be the baseline for activities C and D. The purpose of activity C is to determine the rate at which H2O2 spontaneously decomposes when exposed to room temperatures and ambient light for 24 hours. The purpose of activity D is to determine the rate at which catalase decomposes H2O2. After adding H2SO4 for different time lashes, etc., the resulting data will be graphed at which the catalase decomposed by catalase.…

    3) The experimental system of this lab is, first to set up the logger pro pressure measuring device to the computer. After the device is set up completely, set up the logger pro software that will collect the data for this lab. After both of these steps are complete, get a clean test tube with 1.5 mL of H2O, 1.5mL of H2O2 and then add drops of enzyme needed. Collect data by plugging the black “plug” into the test tube and press “start” on the logger pro software. Continue collecting data with variety of different conditions.…

We dispensed each fluid into the small test tubes by using the correct amounts on the given chart. The test tubes were tested for temperature by placing it in hot water baths at various temperatures. Test tubes 2 & 3 were placed in a beaker at room temperature, 22°C. The rest of the test tubes were placed in hot bathes with a waiting period of ten minute interval. We placed test tubes 4 & 5 in 50°C, 6 & 7 at 40°C, 10 & 11 in 60°C, 12 & 13 in 70°C, and 14 & 15 in 80°C. We placed a thermometer on the test tubes…

Esters are known for their pleasant smells such as perfumes and artificial flavorings in contained labs. They are formed when a carboxylic acid reacts with alcohol and a strong acid such as a catalyst called sulfuric acid (H2SO4) for this lab. The structural formula for esters can be represented as R-COO-R’. The R and R’ symbolizes different alkyl groups that can be combined to the ester. When naming an ester the first part comes from the alcohol followed by the name of the carboxylic acid. Synthesizing for a complete reaction involves a strong acid which makes the esters a final product.…

First we marked 3 test tubes with the wax pencil at 1cm and 5 cm levels and also numbered them 1,2 and 3. Second we filled each test tube with catalase to the 1 cm mark. Next test tube 1 was was placed in refrigerator, test tube 2 in the refrigerator incubator and test tube 3 in boiling water. We let them sit for 15 mins. After the 15 mins was up we removed each test tube one at a time and filled each one with hydrogen peroxide to the 5 cm mark. The tubes were swirled to mix them. We then waited 20 sec.…
