1. What organisms have been successfully decellularized?
Rat, rabbit, and pig livers have been successfully decellularized. Mouse, rat, goat, sheep, pig, non-human primate and human lung have also been successfully decellularized

2. Which ECM proteins have been found to be found in less amounts (or even damaged) in decellularized lung tissue? What might be the consequence of such alterations from normal tissue?
Collagen Type I, IV, elastin and, fibronectin. With the alteration of the normal tissues the lung could lack key qualities that allow the lung to complete its function.

3. Which cells/cell lines have been successful in recellularization? Has the process been 100% successful?
The lung has partially been recellurized. The alveoli,

RWV works by growing the cells in a culture that is horizontally rotating filled with media, there is gentle media mixing similar to the intersitium for the lung tissues.

5. What is the purpose of the study?
The purpose of the study is to observe if the RWV could enhance the effectiveness of recellularization as well as if RWV would affect the differentiation of mesenchymal stem cells in lungs that have been decellularized.

6a. Summarize the results from figure 2 in your own words.
Figure 2 represents lungs that have been decellularization and then recellurized in two different manners. The bioreactor provided more abundance in cells than the static condition did. With the bioreactor there was more evenly distribution of the cells. Though in both tests there was cell growth mostly in the alveoli, with minor development of the larger airways. There were slight morphological differences between static and bioreactor development.

6b. Do you agree with the conclusions from figure 2? Why or why not?
Yes. The bioreactive lungs seem to have provided the most promising results to maybe be able to fully complete Decellularization and Recellularization in

Do you agree with the conclusions from figure 3? Why or why not?
Yes. With the elevated amounts of RNA there could be the potential for cells to be fully made and produce functioning organs. As seen in previous experiments bioreactive has been producing the more favorable results, and as in this figure bioreactive shows better results as expected.

8a. Summarize the results from figure 4 in your own words.
During the 14-day measurement the MCS’s cells show more staining for Annexin V in the static lungs than the bioreactive as well increased PCNA for the bioreactive lungs. C10 contained higher levels of Annexin V during the 11-day measurement for static lungs, less were present in the bioreactive lungs.

8b. Do you agree with the conclusions? Why or why not?
I kind of agree with the conclusion. The PCNA are showing cell growth while the annexin V shows apoptosis. There is evidence of both almost equally in some sections. Where it seems you would need more growth than cell death. I do agree with the fact that the static lungs showed much more apoptosis than the bioreactor, allowing them to say the bioreactor provided good

They are the basic functional units of lungs and have simple squamous epithelial cells. Alveoli cover about 60-70m.     Human embryonic stem cells are pluripotent stem cells derived from the inner cell mass of the blastocyst, an early-stage embryo. To culture these stem cells under controlled environments, one first must have a growth medium. This growth medium is made with DMEM/F12, which is cell culture medium, a 20 percent Knockout Serum Replacement, 100 μM β-Mercaptoethanol, 1 percent non-essential Amino Acid, and a Basic Fibroblast Growth Factor. This medium is then changed out everyday. This is done threw aspiration of the growth medium. One then can add 2mL of fresh medium per one well of a six well culture plate. Then you can passage the cells every 5 days. Cell passaging or splitting is a technique that enables an individual to keep cells alive and growing under cultured conditions for extended periods of time. In order to do this, you must remove the culture medium and add ½ mL of collagenase IV to each well of the 6 well plates. Then, the cells can incubate for 2-4 minutes. You then gently aspirate the collagenase and gently wash the cells with PBS. You then remove the PBS from the well and add 1mL of fresh culture medium to each well. Then pipette the medium up and down in well to detach colonies completely from the surface. You also manually scrape the tissue culture plate with the pipette tip. Finally, you take the prepared culture plate with feeder cells and split the human embryonic stem cells at a 1:6 ratio.…
