Microbiology Unknown #94 The purpose of this report was to identify an unknown bacterium by applying methods that were previously learned and conducted in microbiology lab class. It is possible to identify a mixed culture by running experiments on the unknown bacteria and taking what we already know and applying it to the results. It helps to slowly eliminate any bacteria that do not correspond with the results of certain tests. An unknown test tube labeled 94 was given to me b my instructor.
The first procedure performed was an isolation of my unknown bacteria with the goal of obtaining a pure culture. This was done by streaking the unknown onto a nutrient agar plate using the streak method. The plates were incubated for two days and the bacterium was able to grow. I studied the bacteria based of its physical characteristics of how it grew on the agar. I began my quest by conducting a Gram stain on my bacteria. I prepared a smear by placing a drop of water onto the center of a slide and then removed a loop full of bacteria to place within the water droplet. I use the aseptic treatment while doing this making sure to flame the inoculating loop before and after transferring bacteria. It was important to get this done correctly because the test determines the stain as well as the shape of the cells. After looking at my stain under the microscopic I came to the conclusion that my bacteria was a positive cocci. After the gram stain I performed specific tests based of those results of the stain to determine what my unknown could be. The first test I did was the catalase test. It had a positive reaction meaning the bacteria produced the enzyme catalase. This helped me narrow my unknown down to Micrococcus and Staphylococcus. I inoculated my unknown onto a mannitol salt agar. The growth and color change to yellow indicated mannitol fermentation because acid was produced.     In this lab experiment we did several test to determine what our unknown bacteria was. To determine this we recorded the results of how the bacteria reacted to different media. Depending on the results of each test we could narrow down the different bacteria to determine what our unknown is.     Since it was gram positive, I knew my organism had a thick peptidoglycan and no outer membrane. After finding out my organism B was gram positive, I did the catalase test on it. After adding the two drops of H202 and seeing bubbles, I ruled out Streptococcus lactis. My organism did have the catalase enzyme; meaning it was either Bacillus subtilis or Staphylococcus. To distinguish the two, I performed the simmons citrate test on them. Since the tube stayed green I confirmed that my organism was Staphylococcus. My organism B could not degrade citrate, so that meant my organism was not Bacillus subtilis. In conclusion, my unknown organisms were Staphylococcus and Enterobacter aerogenes.…

    An unknown bacterium was handed out by Dr. Honer. The appropriate tests were prepared and applied. The first procedure that was done was the gram stain. Under a microscope, if the gram stain is purple, the bacterium is gram positive, if the stain is red, it is gram negative. The next test was the fermentation tests for glucose, sucrose and lactose. If an organism is able to ferment the carbohydrate, there will be acid produced, which will turn the broth yellow. Homofermentation shows that only one product was produced-acid. Other organisms will carry out heterofermentation, the presence of acid and gas. If the organism is unable to ferment the sugar in the tube, there will be no change in the orange/red color of the broth and will be considered alkaline. The following tests that I performed were tests for methyl red and vogues proskauer in SIM media. A positive methyl red test indicates a mixed acid fermentation and would turn the organism’s broth red, if no color presents, it is negative. A positive vogues proskauer test indicates fermentation to neutral products and will turn the broth red, if no color presents, it is also negative. Stab inoculating the SIM media for these tests will also show if the organism produces sulfur or indole and if it is motile. If sulfur is present, then the medium will be black, if it produces indole, the medium will be red, and if it is motile, there will be growth throughout the tube. I also grew the organism on MacConkey agar. After inoculation, visible colonies may or may not be present.…
