Unknown Number 94
Unknown Results Streptococcus salivarius
The first step in my unknown identification was to carry out a gram stain to observe the cell shape and arrangement. My unknown turned out to be gram positive bacteria cocci shaped with long chain cell arrangement. The unknown was gram positive because the cells stained blue-violet ,this indicate that my unknown has ability to retain crystal violet-iodine after alcohol application on the cells which is a gram positive characteristic caused by more peptidoglycan than gram negative organisms on cell walls . To observe the cell morphology I used a light microscope oil immersion for a total magnification of 10x 100x =1000x as mentioned earlier the cells were cocci with long chain arrangement.
After the gram stain test I carried out a catalase test. This test was carried out to investigate if my unknown was streptococci (negative) or staphylococci (positive) and or micrococci (positive). The catalase test result indicated that my unknown was negative because no bubbles formed when I placed a loop of the organism into hydrogen peroxide.
The next step was to examine a blood agar plate to examine the colonial morphology of hemolysis , my organism produced gamma hemolysis on the blood agar because there was no hemolysis on the plate after 48 hours incubation period.
After examining my unknown for hemolysis I set a series of five experiments: Bacitracin SX CAMP
Enterococcosel
6.5% NaCl
My unkown teste to be variable to both bacitracin and enterococcosel test. Bacitacin test was carried out to determine if bacitracin antibiotic would prevent the synthesis of peptidoglycan on my unknown organism cell wall, sensitivity of bacitrin would be seen by the formation any zone of inhibition around the bacitracin disk meaning growth was not possible. My unknown was variable to bacitracin because there was growth on one side of the disk with the other side showing no growth at
