II.Results and Discussion
A.Isolation of Agrobacterium tumefaciens:
Six bacterial colonies were observed and screened on the selective medium. The bacterial colonies was able to characterized after 72 hours of incubation, bacterial colonies were visible with naked eyes on YEMA plates. The colonies appeared were medium sized bubble shaped, round, regular, sticky and white coloured colonies were observed. From these initial results, the isolated bacteria were tentatively identified as Agrobacterium strains.
B.Characterization of Agrobacterium tumefaciens:
Biochemical tests: biochemical features of the selected isolates were given in the table 1. The Gram reaction indicates selected isolated are Gram negative. All the six isolated are positive

The negative control was E.Coli and positive control was identified Agarobacterium strain.

D.Molecular analysis on protein profiling of isolates:
The protein sample of six isolated was showed the distinguished protein bands in the range of 66KDa to 14KDa. The molecular weight sizing data showed that the all the six isolates were diverse Table 2. The data generated on the polymorphism study was confirmed the diversity in the isolates table 3.

In previous studies effective protection against tetracyclin was obtained (18) and also confirms that the Agrobacterium tumefaciens have the sensitivity to many antibiotics including tetracyclin and kanamycin (19). Antibiotic resistance means the abililty of a microorganism to withstand the effects of an antibiotics(20). The isolated were showed sensitivity towards ampicillin, chloramphenicol, penicillin, streptomycin, sulphatriad confirms the differnce in response between the isolates should reflect natural

    The streak for isolation was chosen to see the formation of the different colonies and to see if the bacteria would cause growth on the plate. After observing the plate, it could be seen that the unknown had growth and formation of colonies. The Gram stain was chosen next to determine if the unknown was gram-negative or gram-positive. After looking at the gram stain under the microscope with the oil immersion it could be determined that the unknown was gram-negative due to the fact that the bacteria was pink in color and rod shaped. After determining the unknown was gram-negative, other bacteria’s were able to be eliminated from the options. The Triple Sugar Iron Test was chosen next because the unknown could be tested for glucose fermentation, lactose fermentation, sucrose fermentation and sulfur reduction. If the organism is able to ferment glucose and lactose and/or sucrose it will turn the medium yellow throughout (Leboffe & Burton 329). When the Triple Sugar Iron Test was complete, it showed a yellow butt, yellow slant and gas production. This determined that the unknown ferments glucose and one or both of the other sugars. The gas was produced by fermentation. The color change in the agar brought the unknown down to two options of unknowns, Escherichia coli or Hafnia alvei. After both plates were properly inoculated and incubated at 37C for 24- 48 hours, and two separate morphologies were then observed and recorded, unknown A and unknown B, a Gram stain was then performed for each unknown. After determining the Gram reaction, alternate staining procedures were performed as needed. Unknown A required further endospore staining, whereas unknown B required no further staining procedures.
