Objective of experiment * The objective of the experiment is to assess the effect of different pH conditions on enzyme activity.
Hypothesis
* I feel the pH affected the enzyme activity to produce product.
Prediction
* If the pH affected the enzyme activity to produce product, then rate of reaction will change.
Procedure
* The procedure that we followed was to take two test tubes and label one pH2 and one pH10 and incubate 1 ml of each solution for 30 minutes. Then after 30 minutes we added a) 3.5 ml buffer b) 0.5 ml ONPG (substrate) c) 0.5 ml of the dilution that was incubated with the buffer (enzyme)
Then we allowed the reactions to proceed for the 19 seconds, which we used in activity 2 for the pH7. Then we stopped it with 0.5 ml sodium carbonate. Then put it in the sec 20 to determine the absorbance of each solution. The concentration enzyme was obtained by with the standard curve as attached. The rate of reaction is calculated by concentration/time. * The dependent variable is the rate of reaction. The independent variable is pH levels. The controlled variable is temperature in the room and the enzyme concentration. Conclusion * The conclusion of this experiment was supported by the rate. The results between the pH 2 and Ph10 decreased compared to the pH 7 found in activity #2. The absorbance found is that it does procedure product. My results agree with my

    Conclusion: This experiment was to test if increased amount of pH will affect the rate of reaction in an enzyme catalyzed reaction. The hypothesis was correct in stating that the increased pH will increase the rate of reaction. In Test one the qualitative color was only at nine after five minutes of timing the reaction. In the next experiment the qualitative color was at ten, for some, before the four minute mark. This being the case, the experiment does not support our hypothesis. The graph above shows the rate of reaction. Within the first three minutes of the reaction drops at a very rapid rate. Then, the next two minutes the rate of reaction has a huge increase. After this wave the reaction steadily decreases until it halts. Since, at the beginning of the reaction the reaction dropped, the color changed very slowly. With the increase the color changed very dramatically. In conclusion, increasing amount pH to the reaction did help increase the time it took to finish.…

    After performing our tests we came to the following conclusions. Increase in enzyme concentration speeds up the enzymatic rate. The reason for this is when you increase the amount of enzymes (enzyme concentration) then you have more enzymes breaking substrates. Since there are more enzymes more substrates will be broken down than if you have less enzymes. Decrease of temperature leads to decrease of enzymatic rate. This is because when you decrease temperature, you decrease energy. Since energy is decreased the speed of the enzymatic rate goes down. Decrease in substrate concentration leads to decrease in enzymatic rate. When the number of substrates goes down the enzymatic rate goes down because since there is less substrate to break down.
