Objective of experiment * The objective of the experiment is to assess the effect of different pH conditions on enzyme activity.
Hypothesis
* I feel the pH affected the enzyme activity to produce product.
Prediction
* If the pH affected the enzyme activity to produce product, then rate of reaction will change.
Procedure
* The procedure that we followed was to take two test tubes and label one pH2 and one pH10 and incubate 1 ml of each solution for 30 minutes. Then after 30 minutes we added a) 3.5 ml buffer b) 0.5 ml ONPG (substrate) c) 0.5 ml of the dilution that was incubated with the buffer (enzyme)
Then we allowed the reactions to proceed for the 19 seconds, which we used in activity 2 for the pH7. Then we stopped it with 0.5 ml sodium carbonate. Then put it in the sec 20 to determine the absorbance of each solution. The concentration enzyme was obtained by with the standard curve as attached. The rate of reaction is calculated by concentration/time. * The dependent variable is the rate of reaction. The independent variable is pH levels. The controlled variable is temperature in the room and the enzyme concentration.     The optimum pH for the enzyme acid phosphatase was predicted to be within acidic regions and the results obtained showed that the optimum pH was about 5.5 see fig.10. It had the highest absorbance value, meaning it had the most PNP in the tube in the given time and thus the fastest rate of reaction. A change in pH changes the shape of the active site of the enzyme. The bonds within the active site of the enzymes are polar, this means that they are extremely sensitive to ions. The decrease in pH increases the concentration of H+ ions in the solutions, these interact with the polar bonds in the enzymes structure to form individual bonds. This disrupts the shape of the active site and thus the substrate PNPP is no longer complementary to the enzyme’s active site. So no Enzyme substrate complexes can be formed and the rate of reaction drops. The same thing happens when there are extra OH- ions in the mixture. The pH in our cells must be extremely specific and buffered in order to prevent changes in pH and the denaturing of these enzymes.     Conclusion: This experiment was to test if increased amount of pH will affect the rate of reaction in an enzyme catalyzed reaction. The hypothesis was correct in stating that the increased pH will increase the rate of reaction. In Test one the qualitative color was only at nine after five minutes of timing the reaction. In the next experiment the qualitative color was at ten, for some, before the four minute mark. This being the case, the experiment does not support our hypothesis.     From the graphs, it is evident that an increase in both catalase concentration and substrate concentration resulted in a higher rate of reaction or, as observed in the kPa graphs, a higher volume of O2(g) formed at the end of the 5 minute trial. Interestingly, it should also be noted, as it was mentioned in the Figure 2, that the trend for the 6mL of 3% H2O2(aq) was more of a linear trend than an exponential decay, steadily rising until the end of the 5 minute trial. From this, it can be inferred that this trial experienced a saturation point for the enzyme (catalase) to substrate (H2O2(aq)) concentration ratio. Thus, to truly understand this, the trial time period should be extended insofar that a declination in the rate of the reaction can be observed with multiple trials. If the trends of the independent trials coincide with one another, then it is plausible that a saturation point may have been a factor of the linear-like trend. This case will be further discussed in one of the five major factors that influence enzyme activity: enzyme concentration, substrate concentration, pH, metal salts, and temperature.
