Unknown Microorganism. 17 Lab Report
In order to properly performed this test, the initially slant for unknown microorganism #17 is important. For this lab, two identical slants are used for two reasons. Firstly, the slant can be used to make sure that there is no contamination from the Nutrient Agar plate. Secondly, the second slant will become a stock culture to prevent the shortage of slants during performing the series of tests.
Kliger’s Iron Agar tests can be used to determine multiple characteristics of unknown microorganism #17. Kliger’s Iron Agar slants also contains a pH indicator, phenol red, which can be used to test the presence of fermentation. If there is a glucose or lactose fermentation, the acid will be produced, and the color will change from red to yellow. If there is only glucose fermentation, the slant will have a yellow butt. If there are both glucose and lactose fermentation, there will be a yellow slant and butt. Kliger’s Iron Agar slants is also used to check the presence of hydrogen sulfide. If hydrogen sulfide reacts with ferrous sulfate, the slant will have a black butt, and the test result is positive. Otherwise, if the slant has a yellow butt, the test result is negative. This agar also contains a pH indicator, bromothymol blue, which can be used to identify the presence of oxaloacetate and pyruvate (1). If the slant changes to color blue, which indicates that the microorganism is able to produce citrate, and the test result is positive. Otherwise, if the slant remains a green color, which indicates that the microorganism is not able to produce citrate, and the test result is negative.     An unknown bacterium was handed out by Dr. Honer. The appropriate tests were prepared and applied. The first procedure that was done was the gram stain. Under a microscope, if the gram stain is purple, the bacterium is gram positive, if the stain is red, it is gram negative. The next test was the fermentation tests for glucose, sucrose and lactose. If an organism is able to ferment the carbohydrate, there will be acid produced, which will turn the broth yellow. Homofermentation shows that only one product was produced-acid. Other organisms will carry out heterofermentation, the presence of acid and gas. If the organism is unable to ferment the sugar in the tube, there will be no change in the orange/red color of the broth and will be considered alkaline. The following tests that I performed were tests for methyl red and vogues proskauer in SIM media. A positive methyl red test indicates a mixed acid fermentation and would turn the organism’s broth red, if no color presents, it is negative. A positive vogues proskauer test indicates fermentation to neutral products and will turn the broth red, if no color presents, it is also negative. Stab inoculating the SIM media for these tests will also show if the organism produces sulfur or indole and if it is motile. If sulfur is present, then the medium will be black, if it produces indole, the medium will be red, and if it is motile, there will be growth throughout the tube. I also grew the organism on MacConkey agar. After inoculation, visible colonies may or may not be present.…

    My dichotomous key was the “key” thing to finding my unknown out. After the gram staining of the gram negative, the O-F test was the first test to start my process off; it produced yellow through out both tubes. This meaning it is a fermentor of glucose and can be oxidative of glucose. It is also considered O-F because the mineral oil is added to one to promote an anaerobic growth and fermentation, while the one w/o oil is for oxidation and aerobic growth and it grew well in both. After finding out I had a fermentor the second thing was to do perform an Indole production experiment in order to cut my chances of finding my unknown in half. The Indole production test was recorded as negative. It is observed as negative because after adding the Kovac’s reagent there was no color change. Kovac’s reagent added to the tube reacts with DMABA and with any Indole present and produces a compound that turns the reagent layer pink. No color change for an Indole Production means tryptophan is not broken down into Indole and Pyruvate. After finding out I was negative for Indole, looking at my key my next test was Phenol Red Arabinose which also put me at a four to two ratio for finding out my unknown. After performing this test I found out this test is also negative. I recorded this test as negative because there was no color change in the tube meaning no reaction/no fermentation. Phenol Red Broth is a differential medium which also has a carbohydrate added. Phenol red and peptone are included in the medium as ph indicators; an indicator of gas production called Durham tube is also added. When the pH is raised it turns the tube pink, when lowered it turns it yellow and a bubble would mean gas production.     Both media are selective and favor the growth of gram-negative bacteria. EMB contains eosin and methylene blue while SS contains bile salts and brilliant green dye, all of which inhibit the growth of gram-positive bacteria. As the name suggests, SS agar should theoretically only allow the growth of Salmonella and Shigella. In fact, many gram-negative bacteria cannot grow on SS agar. However, some enteric bacteria are able to grow on this medium, due to shared characteristics with Salmonella and Shigella, though their proliferation is limited. Both EMB and SS agars are also differential as they contain lactose, which causes indicator dyes in the media to change color in response to increased acidity when it is fermented.
