Figure 1

A. B. C. D.

Figure 1. Confocal images at 400x magnification of HeLa cells to locate GFP activity. HeLa cells were (A) tagged with Green Fluorescent Protein (GFP) (B) labeled with GFP and 14.13 µl Dexamethasone (C) tagged with GFP fused to the glucocorticoid binding domain (GBD) and (D) the subcellular localization of GFP-GBD in the presence of 14.13 µl Dexamethasone.

HeLa cells were transfected with a vector plasmid containing either GFP (A,B) or GFP-GBD (B,C). Following incubation for two days in 1 ml of complete media, the cells containing 0.45 ml of transfection complexes were washed with 1 ml HBSS to remove any

Figures (B) and (D) included 14.13 µl of dexamethasone whereas figure (C) was without dexamethasone.

A plasmid vector pRK5 has been transfected into HeLa cells. 125 ng of 210 µg/ml pRK5-GFP DNA and 100 µg/ml pRK5-GBD-GFP DNA were added to the Buffer EC in different wells at the same ratio, 0.595 µl and 1.25 µl, respectively. Two of the wells contained GFP DNA whereas the other two wells contained GFP-GBD DNA. All four of the wells were condensed with 1 µl of an

    1. Some potatoes increased in mass while others decreased due to osmosis. Potatoes that had a higher concentration increased in mass because water passed through the membrane into the potato in an effort to dilute it. Potatoes that had a lower concentration than the solution decreased in mass because water flowed through the membrane into the solution trying to dilute the solution. The water moves to the higher concentration to dilute it/maintain homeostasis, increasing or decreasing mass depending on whether it goes in or out.…

For many decades, HeLa cells have helped scientists develop many achievements in vaccination, mapping chromosomes, and even cloning. HeLa derived from the first two letters of the first and last name of Henrietta Lacks. These cells were taken from her in the 1950’s without her or her family’s knowledge. Only twenty years after her death did they discover its’ existence.…

Transformation is the genetic alteration of a cell resulting from the direct uptake of exogenous DNA from its surroundings. By doing this lab, we will answer the essential question: What influences transformation efficiency? Although there are many possible influences, our hypothesis is that plasmid has a positive influence. To test this hypothesis, we will set up an experiment in which we predict that if plasmid is present, then the transformation efficiency will increase.…

Protein-DNA complexes were formed using 1 µg of plasmid pTriEx-1.1 Hygro at different pDNA: protein molar ratios (1:100, 1:200, 1:500, 1:1000, 1:2000, 1:8000) during 10 minutes. The increasing molar ratios were assessed by gel retardation assay on a 0.8% agarose gel and visualized by ethidium bromide staining. For further experiments, all samples were prepared in the 1:8000 pDNA:protein molar ratio with 1…

Human Embryonic Kidney(HEK) 293 cells were transiently transfected with pEGFP-N1(Clontech, Palo Alto, CA, encoding enhanced GFP as an indicator of transfection) and pEGFP-LeIF plasmids cloned at previous studies(Ghafarifar and et al, in press) by Calcium phosphate and non- tranfected cells was used as negative control. briefly, Hek-293 cells were grown at 37°C in Dulbecco's Modified Eagle Medium (DMEM, Gibco) supplemented with 10% Fetal Bovine Serum (FBS, Gibco) and 5% CO2. Log-phase cells were employed for transfection experiments. Briefly, 24 h before transfection, cells were trypsinated and spilited into a 6-well plate (about 3 × 104 cells/well). The cells reached 40-60% confluence the day of transfection. 2-3h prior to transfection, culture…

The correct insert size was determined by Colony PCR. Plasmids from clones containing the correct-sized inserts were isolated and the MCS was sequenced using both a forward and a reverse primer (HT7 For and Flexi R). During cloning an extra GTT, which encodes for a valine residue in the proteins' primary structures, was inserted immediately prior to each stop codon. As a universal stop codon TAA was used, replacing other stop codons if present. This was mainly done to gain maximum flexibility during cloning as the Flexi vector system enables the direct transfer to other vectors with different tags. A GTT is mandatory to later transfer the gene of interest to a vector encoding a C-terminal tag.…

As shown in Fig. 5, the results point out a significant increased in internucleosomal DNA fragmentation of HeLa and HepG2 cells. The DNA isolated from cells treated with Pierisin-6 (2 and 25 ng/ml, respectively) concentration were incubated for 48 h and subjected to agarose gel electrophoresis. DNA ladder characteristic of apoptosis were observed in both cell lines (Fig. 5). No DNA fragmentation occurred in the untreated cells.…

This study’s objectives are first to verify the direct proportionality of acceleration and net force if the mass of the body is constant and then to verify the inverse proportionality of acceleration and mass if the net force is constant. With the use of, mainly, the dynamics cart and the weight hanger, certain variables such as the mass on the cart and the mass on the hanger which is the net force were manipulated, controlled or changed in order to illustrate Newton’s second law of motion. However, through the process, some sources of error were expected that makes the results not a hundred percent accurate.…
