Reviewing Your Knowledge

2. A pigment solution contains compound A with 4 polar groups and compound B with 2 polar groups. You plan to separate these compounds using paper chromatography with a non-polar solvent. Predict the location of the two bands relative to the solvent front. Explain your answer. Since compound A is probably more polar than compound B, compound B will move further in the chromatography. Since the solvent is non-polar, the less polar compound will be more soluble in the solvent than the more polar one.

Applying Your Knowledge

2. In response to shortened day length and cool temperatures in the fall, many trees begin a period of senescence when the breakdown of chlorophyll exceeds a chlorophyll production. The leaves of these trees appear to change to yellow and oranges. Using your knowledge of photosynthetic pigments, explain the source of these yellow-orange hues.
The carotenoids, which are the pigments that reflect in the yellow to red spectra and absorb blue light. There are some, functioning as secondary pigments involved in photosynthesis, that is unmasked once the chlorophyll content diminishes.

3. Duckweed is a tiny floating aquatic plant that can reproduce so rapidly that it can completely cover the surface of small fish hatchery ponds. One proposal to limit the growth of this invasive plant is to use colored panels to shield the ponds, inhibit photosynthesis, and kill this pest. What color filters would you order for the panels in this project? What wavelengths of light will be absorbed by theses filters? So you think this proposal will be effective in killing the duckweed?
Primary photosynthesis occurs in the red and blue regions of the visible light spectrum. By inhibiting light passage for wavelengths of 700-635nm and 490-450nm, most photosynthesis would become inefficient. This would most likely kill the pest as well as most other primary photosynthetic life.

In order to experiment with distinct components of a mixture, they must be first separated so they can be observed individually. This is accomplished in this lab by the technique called thin layer chromatography (TLC). TLC involves a stationary phase, which the TLC plates as well as a mobile phase, which could be one of two solvents used: ethanol-acetone for TLC. Dyes in a sample separate consequently because of their unique polarities. As a result, nonpolar substances travel further than polar substances in this process. The separation of a mixture into its components by TLC transpires because the distinctive components of a mixture interact to different extents with the stationary and mobile phases.…

Pigments extracted from different greens have different polarities and may be different colors. Mixed pigments can be separated using chromatography paper. Chromatography paper is able to separate mixed pigments due to their polarity and solubility. Pigments of chlorophyll a, chlorophyll b and beta carotene will be separated on chromatography paper because each has its own polarity and solubility, which results in different distance traveled up the paper. Beta carotene is non-polar so it travels the highest distance, followed by chlorophyll a. Chlorophyll b is the most polar; therefore, it travels the shortest distance. The separated pigments on the chromatography paper can be eluted in acetone and absorbance spectrum is determined using spectrophotometer. Spectrophotometer produces a graph of the absorbance spectrum which shows the wavelengths (in nm) that are absorbed by the pigment and that are reflected. The reflected wavelength of chlorophylls a and b reflect the wavelengths of l light that are associated with green, which gives them their color. Beta carotene reflects the wavelengths that are associated with orange color, and that explains its color.…

Purpose: Unknown mixtures will be separated by means of chromatography in which the mixture will be passed in a solution through a medium leaving behind components of the mixture at different rates, therefore, different spots on the absorbing substance. This will help determine the identity of unknown mixtures. The spot colors on the strip of filter paper and the Rf values of the unknown samples will be compared to those of known samples. To find the position of the spots on the strip of paper, we will use the Retention factor equation: Rf = D/L= (distance component moves)/(distance solvent moves)…

In plants, the pigments in chlorophyll are used to absorb the light energy from the sun to aid in the process of photosynthesis (Biology 102 Lab 7). Specific wavelengths are assimilated into the pigments; however, not all wavelengths will be absorbed into the plant. The wavelengths not absorbed will reflect off of the plant and act as the visual representation of a plant’s color, thus resulting in the green wavelength not being absorbed in the majority of the plant (Book citation). Furthermore, plants must have an increased pigment concentration in order to absorb the greatest amount of light possible (Book citation). The lab suggests that the control test tube will be able to absorb the wavelengths it wants to without a struggle because it will not have any filter hindering the plant’s process of photosynthesis.…

Chlorophyll a absorbed the most light across all areas of the spectrum from a variety of colors. Chlorophyll b mainly absorbed blue and violet light, but also picks up on some red light; whereas xanthophyll absorbs approximately the same amount of blue and violet light as chlorophyll b, albeit without the addition of red light. The trend of the total amount of light absorbed by all pigments most closely matches that of chlorophyll a because it was the most active in light…

I picked the colors for my experiment, by asking myself some questions. The first question I thought about was: why are most plants green? The colors you see are the colors that are reflected from the plants surface, the other colors are absorbed by the plant. This tells me that light in the green range is not absorbed by the plant. That lead to my next question: What if that was the only color available to the plant?…
