The Expression and Purification of Recombinant Green Fluorescent Protein (rGFP) From E. coli strain, BL21(DE3), Using Ni2+-Agarose Affinity Chromatography

Abstract: The purpose of these series of experiments was to express and purify recombinant Green Fluorescent Protein (rGFP) from the E. coli strain, BL21(DE3) by beginning with its purification via a Ni2+-agarose affinity chromatography column. The His6 tag of the rGFP bound to the Ni2+-agarose column and washes and elutions were obtained, with elution 3 containing the most amount of fluorescence at approximately 12,000 RFUs. From here, a Bradford assay was performed in order to determine how much protein was in each sample and an SDS-PAGE/Coomassie Blue analysis was done to determine the size and purity of rGFP in the elution 3 sample. The sample came out to be about 34 kDa and was about 75% pure. Lastly, a Western Blot was performed with binding of primary and secondary antibodies to prove that the protein of interest, rGFP, had indeed been expressed and purified.

Combined activity/elution Profile of rGFP during washes 1-6 and elutions 1-6 in a Ni2+-agarose column. The graph shows RFU’s (left) and total protein in ug (right) for each wash and elution. Once the rGFP had been purified in the Ni2+-agarose column, a breaking buffer (10mM Tris, pH 8.0; 150mM NaCl) was used to create the washes by pipetting the buffer in 0.5ml increments and collecting each in separate tubes labeling them W1-W6. Then, an elution buffer (10mM Tris, pH 8.0; 150mM NaCl; 300mM imidazole) was collected in increments of 0.5ml and labeled E1-E6. We could measure the amount of RFU’s each sample had using 200ul of each fraction and placing them in a spectrofluorometer. E3 showed to have the greatest amount of fluorescence. Using the Bradford assay, the total protein of each sample was determined by extrapolating points from the standard BSA curve. Both values were scaled

    Natural enzymes are proteins that catalyze biological reactions by lowering the activation energy of the reaction without being altered during the process. The enzyme used in this experiment was the β-galactosidase purified from E. coli. This enzyme hydrolyzes lactose and turns it into galactose and glucose. Since it is difficult to assay the activity of β-galactosidase, we will be using the artificial substrate, o-nitrophenyl-β-galactoside (ONPG) instead of lactose. The protein from the previous purification step limits the amount of protein that can be recovered by the subsequent purification step. In this case, the total protein recovered by 65% cut was 159 mg, but Affinity Chromatography recovered 350 mg of protein. Purification steps cannot gain protein unless protein is added during the purification process, which none was.     Affinity tagging of proteins is an important method for biochemical analyses [4] and has proven effective in further expanding our knowledge of their specific functions. A group of interactive proteins expressed by bacteriophage ʎ known as exo, bet, and gam have shown a unique ability to facilitate homologous recombination [4,5,6, and 7]. We describe the techniques behind the recombineering process that enables us to insert a modified affinity tag with neomycin resistance known as “TAP-neo” in between the Z1 and Z2 fragments of the RBM22 gene. Similar research has shown evidence of successful recombination events [3,5 and 6] that have ultimately led to purification procedures of the affinity tagged fusion protein complexes which permit mass spectrophotometric analysis [1,2 4,5,6,7 and 8]. In the experiment the concentration of Orange G that binds to protein was a lot less than either Ponceau S. or Bromophenol blue. The ligand with the highest affinity to BSA was a little more difficult to decipher. The experiment shows that all of the 2 µL of both Ponceau S. and Bromophenol blue bind to the BSA. However, when 5 µL of the ligand is added the BSA is saturated and cannot bind anymore of the ligand. Platelet-Derived Growth Factor (PDGF) is one of many proteins that regulate cellular growth and division. The PDGF family consists mainly of five different isoforms called as PDGF-A, PDGF-B, PDGF-C and PDGF-D. Three active receptors for PDGF include PDGFR-α, PDGFR-β and PDGFRαβ, have been shown to be specific for the PDGF dimers. PDGF-B becomes active when dimeric and it is recognized by the three PDGF receptors. PDGF has served as a study model for the recognition of the nucleotide sequence of an aptamer. High levels of PDGF are indicative of abnormal cellular growth (conglomerates, cancer). Preliminary clinical studies show that PDGF-B levels can be monitored on the bloodstream and, in contrast from its fellow isoforms, it has the ability to activate each of the PDGF receptors (-α, -β and -αβ) shifting the balance of signaling to favor the transformation pathway. This research aims to express and purify the recombinant human PDGF-B in order to study its properties with a selected aptamer. The expression of the PDGF-B gene, cloned in the expression vector pReceiver-B04, will be controlled with rhamnose in a bacterial system suitable for the RNA T7 polymerase. The recombinant protein will be isolated, purified and characterized by SDS-PAGE and Western blot.
