Introductions: Enzmyes are proteins that catalyze chemical reactions. All living things have catalase present in them. Catalase is a common HYPERLINK "http://en.wikipedia.org/wiki/Enzyme" enzyme found in nearly all living organisms that are exposed to oxygen, where it HYPERLINK "http://en.wikipedia.org/wiki/Catalyst" catalyzes the decomposition of HYPERLINK "http://en.wikipedia.org/wiki/Hydrogen_peroxide" hydrogen peroxide, which is damaging to the cells, into water and HYPERLINK "http://en.wikipedia.org/wiki/Oxygen" oxygen. My investigation is the effect of temperature on the activity of enzyme catalase. In this investigation I wanted to determine the effect that temperature has on the enzyme catalase.

Question: What is the effect of temperature on catalase enzyme activity?

Variables- Independent- Temperature

Dependant- Amount of gas produced -Amount of liquid left in test tube Controlled -Amount of Hydrogen Peroxide -Amount of potato enzyme

Materials -Hot temperature water bath (set at 90 degrees C)
-Cold temperature water bath (set at .5 degrees C)
-Room temperature water bath (set at 15 degrees C)
- Hydrogen Peroxide
- Potato
-Blender
-Cheese cloth
- Water
-Pipette
-5 small test tubes (5.2 ml each)
-5 regular size test tubes
-Paper
-Pencil
-Refrigerator
-Thermometer

Method
Collect all of materials on the materials list
Make Potato Enzyme. To make this take 100 grams of potato and add 200 grams of water. Blend in a blender till liquified. Then filter liquid through 2 layers of cheese cloth. Once complete keep in the refridgerator at 40 degrees.
Set up all water baths to correct temperatures. Set hot water bath at 90 degrees C, Cold water bath at .5 degrees C, and room temperature water bath at 15 degrees C
Set up 5 small clean 5.2 ml test tubes on

The original research question was exploring the effect temperature would have on a yeast catalase reacting with hydrogen peroxide (H2O2). To address the latter question a series of experiments were conducted. The various temperatures experimented with were as follows: 22 degrees Celsius (room temperature), 0 degrees Celsius (freezing), 100 degrees Celsius (boiling), and 37 degrees Celsius. Along with variations in temperature, the experiment involved different amounts of the two solutions being used. Using a spot plate containing 12 wells the different mixtures of hydrogen peroxide and water were as follows: 40 drops of pure hydrogen peroxide (H2O2), 20 drops of tap water and 20 drops of hydrogen peroxide, and 10 drops of hydrogen peroxide and 30 drops of tap water. The results after all experiments combined can be summarized. The reaction between hydrogen peroxide and yeast catalase increases as the temperature increases.     An enzyme is a biological catalyst that speeds up the rate of reaction in certain biological functions. They play a vital role in many aspects of human physiology and are necessary for the functioning of a number of systems, for example in the digestive system to help to break down food. All enzymes have a unique active site that can fit on to a particular molecular arrangement on a target substrate; a substance e.g. carbohydrate, protein, or fat, that the enzyme is designed to breakdown. There are a number of different enzymes in the human body; each type produced specifically to perform a certain role. Enzymes are not themselves destroyed in the reaction to break down a substrate but their effectiveness is reduced under certain adverse environmental conditions. The two most important ones are temperature and PH level; also concentration of enzyme is also a determining factor. Enzyme action is biochemical in nature and, in line with many chemical reactions; it speeds up with an increase in temperature. This would continue until a certain critical temperature is reached where it’s working efficiency drops suddenly. This is due to a deforming or ‘de-naturing’ of the active site leaving the enzyme unable to bond with its substrate. In humans this can have life threatening consequences if the body temperature rises above 40 degrees Celsius. As enzymes normally function in their own particular part of the anatomy, they operate most efficiently in a medium with a specific acid/alkali (PH) balance. If this balance is either too high or too low it’s functioning is reduced and in extreme circumstances this can lead to de-naturing also.…
