Introduction

Ohm’s Law is V = I * R or in some cases I = V / R. The next two laws were established by a German physicist by the name of Gustav Kirchhoff. Kirchhoff’s first law is his voltage law. Kirchhoff’s Voltage Law (KVL) states that around any loop in a circuit, the voltage rises must equal the voltage drops. The next law that Kirchhoff introduced was his current law. Kirchhoff’s Current Law (KCL) states that the total current entering a junction must equal the total current leaving the junction. These laws, however, cannot be proven or tested without the aid of a multimeter. A multimeter is an instrument designed to measure electric current, voltage, and usually resistance, typically over several ranges of value. The multimeter has different programs to measure voltage, current, resistance, etc. produced from one of two types of circuits. The first type of circuit is a series circuit. A series circuit is a circuit in which the components are arranged end to end in such a way that the electric current flows through the first component, through the next component, and so on, until it reaches the source again. In contrast, a parallel circuit is a circuit in which the has more than one resistor and has multiple paths to move along. The main purpose of this lab was to prove the laws of Ohm and Kirchhoff. On another note, being able to take part in this lab taught my partner and I the fundamental skills of constructing series and parallel circuit and using a multimeter to calculate the current and voltage of a circuit. I believe that if we only have the basic materials to conduct electricity (such as resistors, alligator clip, cords, a multimeter, and a power source), then we can still prove the that the laws that Ohm and Kirchhoff established are in fact reliable sources for calculating data regarding certain electrical circuits. I’ve reached this hypothesis because Ohm’s laws and Kirchhoff’s two laws are supposed to be laws used for

Previously our enzyme was purified through anion-exchange chromatography on Q-Sepharose, but to get the enzyme out of the stationary phase NaCl was added to salt it out.     The homogenates provided were made by homogenizing tissues in a sucrose phosphate buffer in a 1:20 ratio. The protein concentration in bovine cells was measured by diluting the homogenate with a 1:5 ratio; 50 microliters of homogenate and 200 microliters of water. Then 5 known protein concentration samples which were 0.4, 0.8, 1.2, 1.6, 2.0 mg/ml of bovine serum were used to determine absorbance with a spectrophotometer. Two additional samples were made; one was blank and the other was for the specific homogenate sample. Then 3 microliters of bradford assay reagent, which indicates the amount of protein present by color, was added to all samples. The spectrophotometer was zeroed at 595 nm. A standard curve was made with the different absorbencies and concentrations. After the linear equation was formed, the unknown sample concentration was determined using the standard curve equation. A Gel Electrophoresis was used to perform a qualitative analysis. The use of 5 microliters of the homogenate was heated to 80 degrees Celsius. Then the homogenate was transferred to a 2-microliter-protein gel sample buffer. Samples loaded on to the gel was run at 100 v and stained with comassie blue; observations were made next lab.     INTRODUCTION There are many reasons for knowing the identity of microorganisms. The reasons range from the knowing the causative agent of a disease to knowing the correct microorganism in order to make antibiotics. This study was done by applying the following methods; OF Glucose, Indole Production, and Malonate Utilization test for the identification of an unknown bacterium. The methods will assist in determining the unknown bacterium found in a 55 year old male that was passing blood and mucous in his stool. The patient had no pain but mild discomfort during bowel movements. The doctor noted inflammation of the lining of the rectum. The doctor removed a tissue sample for examination and referred the patient to a gastroenterologist for a barium xray and colonoscopy. While utilizing the methods stated above, the bacterium found was a facultative anaerobe due to the Gram Negative Rods, fermentation, and the production of indole. The specific Gram Negative Enteric Bacilli was Shigella dysenteriae. There are many reasons for knowing how to control bacterial growth as well as such as antimicrobial drugs, which assist the body’s defenses against pathogens. Microorganisms are capable of causing infection or spoilage. Controlling bacterial growth is important in poultry farm businesses that use equipment to decontaminate processing plant fluids before disposal. During quality control and safety evaluations, bacteria growth that needs to be controlled can be found. While controlling bacterial growth, clusters of Gram Positive Cocci were found. It was found some ways to control bacterial growth were disinfection, sterilization, antisepsis, sanitization, moist and dry heat, filtration, and radiation; just to name a few.
