1. The lab exercise requires that one use a microscope in which a commercially prepared slide of silk fibers is supplied.

What should one do to complete the microscope setup starting from the point in which the microscope is plugged in and placed on the lab bench in front of the user to the point at which the slide is being clamped onto the stage. One can assume that the microscope is set up such that the stage is in its lowest position and that the low power objective is locked into place.

2. a) State the steps one would follow to achieve an efficient and ‘quick’ focus of a prepared slide in which the letter ‘K’ has been mounted. Assume that one has completed all the preparation as per question 1.

b) In order to see the specimen or aspects of the specimen in greater detail one must increase the magnification. Assuming that one has achieved a focused specimen under low power, what is the proper procedure for setting up the microscope to view the specimen under medium power?

The cover slip must be up if there is one. High magnification objective lenses can't focus through a thick glass slide; they must be brought close to the specimen, which is why coverslips are so thin. The stage may be equipped with simple clips (less expensive microscopes), or with some type of slide holder. The slide may require manual positioning, or there may be a mechanical stage (preferred) that allows precise positioning without touching the slide.…
