Propagation of Phalaenopnis Orchids with Cytokinin Paste
Preparation of Keiki Paste
Materials:
Vaseline (Petroleum Jelly) | 44.0g X 3 | Cetyl/ stearyl alcohol | 9.0g X 3 | Paraffin Oil (Liquid Petroleum) | 5.0g X 3 | Tween 40 | 2.0g X 3 | Benzyl Amino Purine (BAP or Benzyl adenine) | 1.2g | Water | 40.0g X 3 | 500ml Beaker | 3 | Metal stirrer | 3 | 55°C water bath | 1 | Weighing boats | 10 | 95% Ethanol | | Weighing Balance | 1 | | | | | | |

Method 1. 44.0g of Vaseline and 9.0g of Stearyl alcohol was melted in a beaker by placing them in a 55°C hot water bath. 2. 5.0g of paraffin oil and 2.0g of Tween 40 was stirred into the molten mixture. 3. 40.0g of distilled water was added and the mixture was stirred vigourously. 4. 0.3g of Benzyl amino purine was dissolved in a minimum amount of 95% ethanol and was stirred into the warm mixture. 5. The final mixture was approximately 100ml and was stored in two 50ml centrifuge tubes. 6. The above steps were repeated with the addition 0.4g and 0.5g of Benzyl amino purine. 7. The paste was stored in the refrigerator.

Application of Keiki Paste on Phalaenopsis Inflorescence Nodes
Materials
Keiki Paste | 0.3g-0.5g BAP | Sterile Scalpel Blade | 1 | Sterilizing torch | 1 | Metal Spatula | 1 | 95% Ethanol | 50ml | Phalaenopsis Plants* | 6 | | | | |
*Plants selected were of approximately the same size. Each plant has 3 inflorescence spikes with height comparable to the others. Each inflorescence spike possesses at least 3 nodes and a maximum of 5 nodes.
Method
1. The scalpel was sterilized by dipping it into 95% ethanol and flamed by the sterilizing torch. 2. The bracts covering the dormant bud were removed by carefully slicing the bottom with the scalpel and peeling it off. 3. The metal spatula was sterilized with 95% ethanol. 4. The spatula was used to apply the keiki paste containing 0.3g BAP onto the dormant buds of the first 2 plants. 5. Step 4 was repeated with keiki
