2. The Rf values were not recorded in lab. However, spot results were recorded. The TLC plates 1-3 showed the presence of starting material (SM) in the crude product (CP) lane. TLC plate 1 was taken after adding the initial addition of 0.25mL of bleach. TLC plate 2 was taken after adding an additional 0.08mL of bleach because plate 1 still showed presence of SM in the CP lane. TLC plate 3 was taken after adding an additional 1mL of bleach for the same reason. However, TLC plate 4 did not show any SM in the CP lane and this was after adding an additional 3mL of bleach. The plate only showed a CP spot, indicating that the reaction had gone to completion.…

The objectives of this experiment were to investigate the activity of enzymes, components that influence the enzyme’s activity, identify an unknown phosphatase, influence of inhibitors, and determine if inhibition is competitive or noncompetitive. A spectrophotometer evaluated the measurement of color change over a period time due to product being formed. Determining unknown phosphatase and effects from different inhibitors were determined by varying the pH and substrate concentrations. The unknown phosphatase analyzed showed it was an acidic phosphatase. Ph 5 had the most enzyme activity and pH 9 presented 1/3 of enzyme activity, while pH of 11 had little activity. The purpose of this lab was to demonstrate an example of a spontaneous reaction and its connection to metabolism; in this case it was osmosis. The procedures that we used to complete the experiment were taking five eggs that had been soaked in a sucrose solution and then placing each one in containers of sucrose that each had different levels of concentration. We also made sure that the hose was under the graduated cylinder in the container of water. Next, the group poured in 40 ml of 1% Hydrogen Peroxide and catalase solution into the gas stopper. Once the reaction started every 5 seconds, we would record how much oxygen was produced for 30 seconds. We repeated that for two trials. We then used a plastic cup to lower the pH of the catalase solution from 6 to 3.5, by adding 1 M of Hydrochloric acid to the solution.
