Membrane permeability of beetroot
Implementation

My aim for this experiment was to find the effect of temperature on the membrane permeability of beetroot. Since I was unfamiliar with this experiment, I first conducted a preliminary test. I considered my preliminary test as my trial for this experiment. My Hypothesis for this experiment was to found out the amount of pigment loss of beetroot when exposed to varying temperatures.

Betalain pigment found in beetroot

My method for this experiment is:-

* Use a cork borer to cut about 3 cores of fresh beetroot approximately 8mm in length.

* Place them with the help of a forcep on a tile and trim so that it is about 1mm in length. While doing this make sure that no pigment is lost from the beetroot. To trim the beetroot cores in equal pieces I use a scalpel and a ruler.

* After doing this I get good amount of beetroot discs that are of 1mm in length.

* My beetroot discs are 1mm in length because the larger the surface area, the more effect would be there for the diffusion rate on the membrane permeability of beetroot.

* Place the beetroot discs in a beaker of cold, running water (at least 2 min). This is done so that the cellular debris and pigment on the cut surface are completely cleaned off.

* Leave them (for about 2minutes) until no more pigment can be seen coming from the cells.

* Next fill the beaker with distilled water and allow it to stand while you continue with preparation. It will be at room temperature by the time you get to the fifth test tube.

* A thermometer can be used to measure the different temperatures of 0°C, 20°C, 40°C, 60°C and 80°C.

* Next set up 5 boiling tubes in a test tube rack.

* This is done so that I'm able to identify each core of beetroot in different temperatures. I also make sure that all my test tubes and boiling tubes are marked as core A, B, C, D and E. I first conduct a preliminary test.

* Place a disc of beetroot core in each of the labelled test tubes and add about 5ml of water. While doing this make

Through secondary research I found that if I were to prevent the structure of the protein from altering in any way, I would need to keep the pH level constant. Subsequently, I decided to will be using distilled water throughout the experiment as it has a pH 7 and is neutral; neither acid nor alkali. By surrounding the beetroot with distilled water the protein will not have been altered, and the concentration gradient will be fairly constant for each of the temperatures also. This is because I will be exposing the beetroot to the surroundings of the same type of water, maintaining a fair test as distilled water may contain less or more impurities as other types of water, for example: tap water. I will have to control the amount of distilled water I use to place the beetroot pieces in and control this by using a measuring cylinder. I will measure 15cm3 of distilled water each time. It is important to keep the amount of water constant, as the dilution will affect the rate of diffusion, therefore affecting the results.…

Once rinsed the samples were each placed into one of 6 numbered test tubes. Test tube 5 was put in the refrigerator to achieve 5C and 6 in the freezer to achieve -5C, and a 30 minutes timer was set. Next 60 ml of water was heated to 70C in a beaker. The sample from test tube 1 was deposited into the beaker and timed for one minute, after which it was removed and placed back in the tube 1. Three squirts from a small pipette of room temperature water were also put in the tube in order to cover the sample. Once the water was added a 20-minute timer was set. At that time the beaker of water was cooled and the process was repeated at 55C for test tube 2, 40C for test tube 3, and 25C for test tube 4 respectively. By this time the 30-minute timer for the cold samples was up, test tubes 5 and 6 were removed from their coolers and three squirts of room temperature water were added to the tube as was done in the other samples and a 20-mintue timer was set. At this time sample 1’s timer sounded, the beet sample was removed from the test tube, but the water was left in order to evaluate its intensity of color. The same was repeated with each of the samples as their timer…
