Allii Sativi Bulbus Pulveratus
Powdered Bawang Bulb

* Allii Sativi bulbus Pulveratus is Allii Sativi Bulbus that has been cut, freeze-dried or dried at a temperature not exceeding 65˚C, and reduced to a powder.
Packaging and Storage – Preserve in well-closed, light-resistant containers in a cool, dry place.
Labeling – Print the Official Title and the National Title of the powdered bulb on the label as directed under General Notices and Requirements Part II.
Botanic characteristic (see Microscopic Examinationfor crude plant Drugs) – Powdered Bawang Bulb is pale yellow, with characteristic aromatic alliaceous odor and taste. Under a microscope, the powder shows swollen parenchyma cells with granular contents; thin epidermal cells; lignified narrow spiral and annular vessels; scattered starch grains; rhomboid crystals of calcium oxalate; sclereids of the epidermis and dry scales.
Identication (Thin Layer Chromatographic Identification Test for Crude Plant Drugs) – Test solution—Transfer 1.0g of dried allii sativi Bulbus Pulveratus to a suitable glass apparatus. Extract with 10mL of a mixture of methanol and water (95:5), using any suitable method as described in Test Solution under Thin Layer Chromatographic Identification Test for Crude Plant Drugs.
The filter is the Test Solution.
Application Volume—5 µL
Plate—Chromatographic silica gel.
Developing solvent system—a mixture of 1-butanol, 1-propanol,acetic acid and water. (30:10:10:10)
Spray reagent—ninhydrin TS.

Procedure—Proceed as directed in thin layer Chromatographic Identification Test for Crude Plant Drugs. Develop the chromatogram in a chamber containing developing solvent system until the solvent front has moved about 15cm from the spot point. Remove the plate from the chromatographic chamber, mark the solvent front and allow to dry. Spray with the Spray reagent and heat in an oven at 105˚C for 5 to 10 minutes. Observe under daylight. Compare the chromatogram of the Test solution with the Reference Thin Layer Chromatogram D; both thin layer chromatograms exhibit similar pattern and spots.
Foreign matter--not more than 2%
Loss on drying--not more than 15%
Alcohol (95% v/v)-Soluble extractives--not more less than 1%
Water-soluble extractives--not less than 5%
Total ash-- not more than 5%

In order to experiment with distinct components of a mixture, they must be first separated so they can be observed individually. This is accomplished in this lab by the technique called thin layer chromatography (TLC). TLC involves a stationary phase, which the TLC plates as well as a mobile phase, which could be one of two solvents used: ethanol-acetone for TLC. Dyes in a sample separate consequently because of their unique polarities. As a result, nonpolar substances travel further than polar substances in this process. The separation of a mixture into its components by TLC transpires because the distinctive components of a mixture interact to different extents with the stationary and mobile phases.…

Separating the components of the extract, is done through the use of Thin Layer Chromatography. It is one of the standard procedures used in many forensic laboratory when analysizing unknown drugs or mixtures (Howlett and Steiner, 2011). Separation of the mixtures occur based on the pH, polarity of its components, solvent and the thin layer stationary phase (Howlett and Steiner, 2011).…

In this experiment thin-layer chromatography (TLC) was used to determine the composition of an unknown mixture of analgesics given five references of AC, ASP, CAF, IBU, SAL, an unknown consisting of a mixture of three of the references, and two unknowns (6,23) containing a single reference compound. The composition of the unknown mixture and two single unknowns were determined by spotting the reference compounds and the unknown compounds on two TLC plates. Placed them in a developing chamber that contained 0.5% acetic acid in ethyl acetate solvent and observed the TLC plates under UV light after the TLC plates were taken out of the developing chamber. The known reference compounds portrayed spots and calculated Rf values that the unknown compounds matched to. This suggests that the composition of the unknown mixture is SAL, and ASP fused with AC and that unknown compounds 6 and 23 are compositions of AC and CAF.…

Flavonoids are an important group of additives that can be defined as pure substances either natural, extracted from raw materials or synthetic. Chromatography is the separation of two or more compounds or ions caused by their molecular interactions with two phases – one moving and one stationary (Weldegirma 2012). Three types of chromatography are used in organic chemistry: Thin Layer Chromatography (TLC), Gas Chromatography (GC), and Column Chromatography (CC). TLC stationary phase is a solid while the mobile phase is liquid. GC stationary phase is liquid and its mobile phase is liquid. CC is one of the most useful methods for the separation and purification of both solids and liquids when carrying out microscale experiments (Weldegirma 2012). CC stationary phase is liquid and mobile phase is gas, the difference between CC and TLC is that CC uses glass while TLC uses silica gel. TLC provides a simultaneous analysis of dozens of samples in a relative short period of time. The major advantage is that its low cost, low solvent consumption and only small amounts of analytes reacquired (Vlajovic 2013). The thin layer stationary phase in TLC requires small sample size and shorter development distances to reveal their separation potential and provide a faster separation, better resolution and the spots are more compact and are favorable for in situ detection (Ligor 2007). In TLC, Rf is defines as the ratio of the distance traveled by the compound to the distance traveled by the solvent:…

Chromatograph method is a method of separating mixtures of two or more compounds. Two phases are important in this method; one that is stationary and one that is moving. Chromatography works on the principle that different compounds will have different solubilities and adsorption to the two different phases between which are to be partitioned. Thin Layer Chromatography (TLC) is a solid-liquid technique. The mixture is observed when it is in two different phase; a solid (stationary phase) and a liquid (moving phase). Thin Layer Chromatography (TLC) is a technique which is sensitive, cheap and fast. A thin layer chromatograph plate is used to identify drug component under UV. It is used to support the identity of a compound in a mixture when the Rf of a compound is compared with the Rf of a known compound.…
