Practical 12: Digestion in Humans

Aim: To distinguish between 5 liquids to come to a conclusion from which region of the alimentary canal it was extracted from.

Hypothesis

In this practical, we have been given 5 unknown solutions hence it is not possible for us without any logical analysis to come up with a conclusion regarding the positioning along the alimentary canal. We cannot come up with a proper answer or predict results without running through the tests.

I though can base my analysis on the following hypothesis:-

The more the starch, the higher up the alimentary canal the liquid is.
The more the reducing sugar, the lower down the alimentary canal the liquid is.
The more the protein, the higher up the alimentary canal the liquid is.
The more the fats, the more the higher up alimentary canal the liquid is.

Description of the appearance of liquids at the beginning:-
D-33: Light White cloudy Emulsion
D-34: Clear transparent liquid

The protein reacts with copper sulphate in alkaline solution to give a deep purple color with an absorption maximum at 550 nm. The sodium hydroxide solution is added to it to create alkaline conditions.
Procedure:
1. Label the five test tubes on the rack; fill each 2cm3 of the liquids.
2. Then fill each will an equal amount of sodium hydroxide.
3. Add copper (II) sulphate drop-by-drop and note chances if any.

Ethanol emulsion test

The ethanol emulsion test is a test for lipids. Lipids is not soluble in water therefore ethanol is used to dissolve it. The lipids dissolved in ethanol is to be then decanted in water. This results in the suspension of small globules of lipids dissolved in ethanol in water such that they both will not mix. Cloudy white emulsion will indicate the presence of lipids. Adding water to the solution results in emulsion of tiny droplets in the water. These droplets reflect light and give a white, opalescent
