Behaviour of cyanobacterial bloom material following coagulation and/or sedimentation

ABSTRACT
The global increase in detection of cyanobacteria and their metabolites has prompted greater emphasis in optimizing water treatment options for their effective removal. In particular, conventional coagulation and clarification processes have been shown to be effective in removing whole cyanobacterial cells, and consequently a majority of the metabolites. However, the resultant cyanobacterial-laden sludge is often not closely monitored in treatment plants. This study has shown that the sludge supernatant derived from the coagulation/sedimentation of a range of natural cyanobacterial blooms contained extremely high concentrations of metabolites (up to
∼8,000 ng L–1 of geosmin and ∼90 μg L–1 of microcystins), which was attributed to cell lysis.
Furthermore, the fate of the metabolites in the sludge supernatants differed with geosmin shown to be biodegradable, while the microcystins were released at different stages with some variants shown to be more persistent. The release and biodegradation of the metabolites followed pseudofirst-order kinetics with rate constants comparable to experiments using laboratory-cultured

    This was compleated by growing the bacteria on a petri dish with agar, gram staining, then examning under a microscope. The bacteria identified was M. Roseus and B. Megaterium. The identity of these bacteria was mostly determined by the positve gram stain results and the shape they where under the microscope. Both of these bacteria where identified as gram positive (as shown in the table previously). This was determined beause of the nonexistant fatty layer in the cell’s cell wall that trapped the purple crystal violate. The second big thing that identified these bacteria was the shape you could see under the microscope. While looking under the oil immersion lense, the B. Megaterium was shaped as a rod, and the M. Roseus was shaped as a circle. The diference between these two was, the rod was a much longer and skinnier shape compared to the circle whitch was smaller and more round than the rod was. The error in this experiment was that during gramstaining the second bacteria. I was able to identify three different bacteria. One was small purple circles (the M. Roseus), another was purple rods (B. Megaterium), and the last one was small pink circles that was not identified. In this lab, one of the problems that was encountered was the bacterias would not grow.     To identify the resulting colonies a procedure called gram stain is used in the identification of bacteria. Bacteria can be gram-positive, gram-negative, or gram-variable. Most bacteria however, are usually gram-positive or gram-negative. Gram-positive and gram-negative bacteria react differently to staining because of the differences in their cell walls. Bacterial cell walls contain a polymer called peptidoglycan. Gram-positive bacteria have more of this polymer than gram-negative bacteria.
