Copper Sulphate On Amylase
My experiment was to find out the effect of changing the copper sulphate concentration on the rate of the hydrolysis of starch to maltose using the enzyme amylase. Copper sulphate was used because it acted as an inhibitor.

Hypothesis, I predicted that the more copper sulphate added to the solution the longer the reaction would take to complete. This was due to the fact that copper sulphate would interfere with the starch for the active sites on the amylase molecules because it is a inhibitor.

Biological Knowledge, Amylase acts as a catalyst and starch is a complex carbohydrate.

The amylase was used to hydrolyse the alternate glycosidic bonds in starch. By doing this it meant that the starch, a polysaccharide was broken down into maltose, a disaccharide. However, copper sulphate would interfere with starch for the active sites and so prevent the formation of enzyme-substrate complexes. Fewer complexes formed means that the time taken to hydrolyse the starch would be greater and the rate of the reaction would be

    We then repeated this in two more test tubes. We put one of the test tubes in ice (0 C) we put another in the water bath (42 C) then we put one in boiling water (100 C). We repeated the same thing for the boiled amylase. We then checked at five minutes to see if the starch had been digested by the amylase. We did this by putting a little of each of the six samples into a spotting tile. We then added iodine solution to this to see if starch was present or not. I could tell if starch was present or not because if the solution went orange it meant that there was no starch present, so all the starch had been digested, if the solution went blue/black it meant that there was starch present, so the starch had not been fully digested. We recorded the results in a table. We then waited another two minutes so that all together the starch solution had been in there seven minutes.     Enzyme inhibitors reduce the rate of a catalysed reaction. They do this by interfering with the enzyme when combining with the substrate. There are two types of known inhibitors.
