Biochemistry Lab Report

Objective: The purpose of this lab is to analyze enzyme activity at pH neutral for acid phosphatase as the colorless p-nitrophenylphosphate (PNPP) substrate concentration changes. We will empirically determine the concentration of production formation, Vmax and Km from absorbance of yellow PNP in the reaction.

This figure indicates a positive linear trend for the concentration of PNP and absorbance.

    Introduction: The biological membranes are composed of phospholipid bilayers, each phospholipid with hydrophilic heads and hydrophobic tails, and proteins. This arrangement of the proteins and lipids produces a selectively permeable membrane.     PNP or paranitrophenol is an organic compound that undergoes changes when the pH is manipulated of the solution. PNP is also an acid/ base indicator. The compounded starts off as internal conjugated ring with the hydroxyl group and the nitro group single bonded to the aromatic ring. This occurs at the pH of 5. At this point the aromatic compound is colorless and is in the ultraviolet range. However, as the pH begins to increase the hydroxyl group deprotonates, leaving the oxygen with a negative 1 charge. The molecule compensates for the negative charge by conforming to a more stable resonance structure. The structure now has the oxygen and the nitrogen for the nitro group double bonded to the ring. The negative charge lies with one the oxygen atom that is a part of the nitro group. This lead to a highly conjugated system. If a system is conjugated the compound shows the presence by giving off a color. In PNP the color change is most visible at the pH of 10.…

    The optimum pH for the enzyme acid phosphatase was predicted to be within acidic regions and the results obtained showed that the optimum pH was about 5.5 see fig.10. It had the highest absorbance value, meaning it had the most PNP in the tube in the given time and thus the fastest rate of reaction. A change in pH changes the shape of the active site of the enzyme. The bonds within the active site of the enzymes are polar, this means that they are extremely sensitive to ions. The decrease in pH increases the concentration of H+ ions in the solutions, these interact with the polar bonds in the enzymes structure to form individual bonds. This disrupts the shape of the active site and thus the substrate PNPP is no longer complementary to the enzyme’s active site. So no Enzyme substrate complexes can be formed and the rate of reaction drops. The same thing happens when there are extra OH- ions in the mixture. The pH in our cells must be extremely specific and buffered in order to prevent changes in pH and the denaturing of these enzymes.
