Laboratory report
Experiment one Acetylation of α-D- glucopyranose

1. Reagents
(1)2.5g(0.014mol) powdered D-glucose
(2)0.5g ZnCl2
(3)12.5ml acetic anhydride
(4)Cold water
(5)120ml Ice water
(6)Methanol

3. Phenomena and data
The reaction in pear shape flask is not vigorously because the amount reflexed solution can even not bring the raw material attached on condenser back to flask. The color of the liquid in reactor is brown, with gentle swirling, glucose at the bottom can plenary react with acetic anhydride. In the ice water, a considerable gelatinous yellow brown solid come from the dark brown solution. The crude product has the weight of 6.92g (crude yield 126.6%). After 4 times recrystallization, the final product is white with light yellow. The most weight product have melting point with 111.4℃, however the M.P. of product after second recrystallization is 93℃
The final product has the weight of 0.35g (yield 6.4%) 4. Discussion (1) Why 4 times of recrystallization is needed
The impurity contains acetic anhydride, acetic acid, Zinc Chloride, beta-D- glucopyranose, not full acetylated product and many unknown impurities. The acetylated products (both full acetylated and partial acetylated) are not soluble; therefore the water wash can only remove that water soluble matter. In the process of recrystallization, little white solid float on solution.

A reflux condenser was assembled using a 25-mL round-bottom flask, hot plate, and aluminum block. It was equipped with a drying tube filled with calcium chloride to control vapors. Approximately 2.5 mL of isopentyl alcohol was massed and placed in the round-bottom flask that contained a stir bar. Using the same graduated cylinder, about 3.5 mL of glacial acetic acid was added to the flask. A calibrated Pasteur pipet was used to add 0.5 mL of concentrated sulfuric acid to the flask and the flask was mixed immediately. The flask was connected to the apparatus and covered with aluminum foil to help retain heat. Using rubber tubing, water was circulated into the lower attachment and out the top attachment and the mixture was brought to a boil. After heating under reflux for 60-75 minutes, the flask was removed from the heating source and left to cool down to room temperature. The reaction mixture from the flask was transferred to a culture tube and 5-mL of water was added. Upon addition of water, careful shaking, and occasional venting, the phases separated and the lower aqueous layer was removed and discarded. Using the same procedure as explained above with water, 2.5 mL of aqueous sodium bicarbonate was added and the lower aqueous layer was again removed and discarded. The same procedure was repeated one last time with 2.5 mL of saturated aqueous sodium chloride and the lower aqueous layer was removed and discarded, leaving behind the crude ester. The crude ester was transferred to…

Also added was 0.45mL water and 0.165mL acetic anhydride and a magnetic spin vane. An air condenser was attached to the vial and it was heated on an aluminum block, with a temperature maintained at 120C. After the solution dissolved, it was heated an extra 20 minutes to allow the reaction to go to completion then cooled back to room temperature. An ice bath was used to encourage crystal growth. The crystals were removed and centrifuged, then placed on a pre-weighed watch glass to dry and weigh. Percent recovery was found. A small sample was set aside for melting…

It is important to note that one sodium hydride can reduce up to four 9-fluorenones because the are four equivalent hydride ions for every sodium borohydrides. To create the alcohol a dilute sulfuric acid is added to release the final product. It is important to minimize contact of sodium borohydride with air so that the reducing agent is not destroyed. The reaction can be observed as successful as 9-fluorenol is bright yellow, whereas 9-fluorenol is a white color. Approximately 100 mg of crude product is obtained through this experiment. Recrystallization occurs, however there is not one pure solvent that allows for recrystallization of 9-fluorenol. Thus, multiple solvents will be added together, water and methanol, the ratio of water to methanol must be determined for this to occur. Therefore, 1 mL of methanol will be added to the crude product in a test tube that is then taken to a boil in a water bath. Once boiling, water is added dropwise until the solution turns cloudy. The water must be added at the same temperature as methanol. Precipitate should form around the droplet of water added that then dissolves. Once the precipitate stops dissolving. At this point if methanol is added it should dissolve, therefore, this point is the correct ratio of water to methanol. However, in this experiment the reaction is stopped once the solution is cloudy and methanol is not added to make the solution…

The solution then gets foggy/cloudy and white snowlike precipitate is floating within the solution. Heat is added along with a stir bar. The stir bar starts stirring at minute 7. It begins to dissolve the white powder as it spins. At minute 26 the solution becomes clear with barely any flakes left. Sulfuric acid is then added and white flakes are formed. More acid is added until the pH paper turns red. The round bottom is filled with white precipitate. The precipitate is put onto a hirsh funnel that uses vacuum filtration to isolate the precipitate. The product is then added to a beaker with D.I water and heated. It’s put through the hirsh funnel . 3.68 grams of salicylic acid is recovered. The melting point is tested to compare to a perfect sample. The melting point is 146C0. The product is dried and added to a beaker with D.I. water and 10mL of acetic anhydride and 20 drops of concentrated sulfuric acid. This product is chilled to produce crystals. Its then filtered using a hirsh filter using vacuum filtration. Recrystallized and filtered one last time. The product is now acetylsalicylic acid…

Procedure: To a 125 mL Erlenmeyer flask containing a mixture of 95% ethanol (6 mL) and acetic acid (5 mL), dissolve acetanilide (7.4 mmol) and sodium bromide (1.8 g). Place the reaction flask in an ice bath (at least 5oC) for 5 minutes (keep this reaction in the hood). Add sodium hypochlorite (8.3 mmol) by addition of household bleach; leave the reaction flask in the ice bath 5 min. After removing the flask from the ice bath and placing a watch glass over the opening, allow the reaction mixture to come to room temperature over 15 minutes. Place the reaction flask into the ice bath and quench the unreacted bromine by addition of both 5 mL of sodium thiosulfate solution (1 g/5 mL) and 5 mL sodium hydroxide solution(1 g/5 mL ); collect the product by suction filtration. Recrystalllize the brominated product from a minimum amount of solvent. You will need to select a solvent from water, 50% ethanol or 95% ethanol. To identify the best solvent, use what you have learned in Organic I Chemistry Lab, hint: think about what properties makes a good recrystallization solvent. Test your recrystallization solvent in test tubes using the tip of a spatula of your compound. Remember to use a water bath to heat organic solvents. NEVER heat organic solvents directly on a hot plate-always use a hot water bath! Dry the product and obtain melting point range (one fast and one slow) and an IR spectrum. Be sure to record observations in your laboratory notebook.…

-The final product was a pasty, weight powder. Even after it was dry, it was still difficult to get a sample into the capillary tube because the product kept sticking to the sides of the tube. This may not be a property of the compound though it’s possible it could be a result of incomplete drying. The melting point range of the product was 246-249 C.…

The volume of 20 mmol of 2-methylcyclohexanol was calculated and transferred into a round-bottom flask. Then 0.7 mL of 85 % phosphoric acid was mixed in. The reaction vessel was clamped to a ring stand over a suitable heat source and an apparatus was assembled for simple distillation using a Hickman still, water cooled condenser, and thermometer. Then a conical vial was weighted and recorded. After turning on the heat source in slow power, it was waited until it started to distillate. The temperature when it began was 58°C. Using a Pasteur pipet, the distillate was regularly transferred into the conical vial until only a yellow solution remained. It wasn’t left to dry completely. The temperature was 75°C when the vessel was taken from the heat source. The distillate was then washed with 1.5 mL of 5 % aqueous sodium bicarbonate. Then the aqueous lower layer was carefully removed. The upper layer was cloudy at first until it started to become clear after a few minutes. The solution was dried through anhydrous calcium chloride. Then it was weighted.…

Experimental: Sodium metal was patted dry to remove any oil and was cat into small pieces. A dry 100cm3 round bottom flask was placed on a cork ring, on a balance and tarred sodium metal (0.6g) was placed into the flask. The flask was then attached to the dry reflux condenser and industrial methylated spirits (IMS, 15cm3) was added. Once all of the sodium has dissolved the solution was cooled to room temperature and the para-acetamidophenol (3.5g) was added. Ethyl iodine (3.0cm3) was slowly introduced to the mixture through the top of the condenser and the resulting in the mixture was boiled at reflux temperature for 20 minutes and was placed in a rotary evaporator to remove any excess solvent. Distilled water (40cm3) was added to the mixture and the flask was placed in ice until crystals were formed. The crude product was filtered using vacuum filtration the filtrates were washed with very little cold water and were left dry as much as possible.…

The three substances, anhydrous diethyl ether, maleic anhydride, and the eucalyptus oil combined a yellow mixture. The mixture produced a very strong scent. After heating the reaction under reflux for 45 min, crystals began to form when reaction was cooled to room temp. The crystals were white when formed and contained a little yellow tent to them. A lot of crystals were formed in the round bottom flask. The crystals were accidently dropped into the ice cold water forming a milky white substance, but this did not harm or contaminate the ending results. After vacuum filtration and being washed with the petroleum ether, the crystals turned pure white.…

Once the ten minutes are up, the solids are then isolated by vacuum filtration, in a set up that includes a Hirsch funnel, and then rinsed with 9.8 mL of hot water and 5.0 m of ice-cold 95% ethanol. Once rinsed, the solids were then recrystallized with a 1:3 ratio of ethanol:acetone; which include 10 mL of ethanol and 20 mL of acetone, giving us a total volume of 30.0 mL. Once the 1:3 ratio of ethanol:acetone was added, the crude produce is then dissolved by heating the solution on hotplate with the heat knob on about 3-4, so as to not overheat the product. Once fully dissolved, the solution is left to cool to room temperature, then placed in an ice-bath for about 20 minutes. Then once again, the solid is isolated through vacuum filtration and a Hirsch funnel. The mixture is rinsed with 10.0 mL of water and 5.0 mL of 95% ethanol.…

Organic compounds that have been synthesized in the laboratory or isolated from natural sources often contain impurities. Recrystallization is a process which removes impurities from solid organic compounds that are solids at room temperature. The solubility of a compound increases with temperature. As the solution cools the solubility of the compound decreases and crystals begin to form and grow.…

Aluminum chloride (AlCl3, 1.45 g) was quickly added to the round-bottom flask by removing the rubber septum, with caution as to minimize exposure of moisture from the air to the reaction mixture. The rubber septum was replaced and the reaction flask was cooled in an ice bath that was prepared in a 100-mL beaker. When AlCl3 was added to the mixture, the mixture began to bubble, changing the color from yellow to a dark purple. Using a plastic pipet, acetic anhydride (0.6 mL) was added to the mixture. The ice bath was removed and the water aspirator was turned on. A hot water bath was prepared by heating 300 mL of…
