Simple staining, gram staining, endospores and special staining and bacterial shape

Experiment 2

Instructor:Dr. Emilia rappocciolo
Student name: Mai ghassan abbas
ID: 1091407

Objective:
To view stained bacteria under a microscope also, recognizing certain bacterial structures and features.

Theory:
It is possible to view unstained live bacteria under the microscope, but this method is uncommon. The method that has been used in this experiment is staining and killing the bacteria. Killing is the step that takes place before staining, it is done by passing the slide over the heat several times, and as a result the bacteria will be attached to the slide. After that staining occurs this is the process that colors the microorganism with a dye that allows us to see certain microbial structures and features.
There are many types of staining; first simple stains its purpose to show the cellular shapes and basic structures. Second, differential stains this stain reacts differently with different types of bacteria so it can be used to distinguish among them and the most common type is the gram stain. Finally special stains they are used to color and isolate specific parts of the microorganism such as, endospores.

Results:
Simple stain:
Bacterial name | Shape of the bacteria |
Esche coli | Coco-bacillus |
Differential stain (gram stain):
Bacterial name | Shape of the bacteria | Color of the staining | Bacterial type |
Bacillus subtilis | Bacillus (rod) | Violet | Gram positive |
Esche coli | Coco-bacillus | Pink | Gram negative |
Special stain (endospore staining):
Bacterial name | Shape of the bacteria | Results |
Bacillus subtilis | Bacillus (rod) | |

Discussion:
Depending on the gram staining, we can divide the bacteria into two major groups, gram negative and gram positive the major difference between them is the composition of their cell... [continues]
