1.Detection of Alkaloids

a.Dragandroff’s test-1ml sample was added to 1ml of dragandroff’s reagent.An orange red precipitate indicates the presence of alkaloids. b.Wagner’s test-1ml of sample solution was added to 2ml of Wagner’s reagent.The formation of reddish brown precipitate indicates the presence of alkaloids. c.Mayer’s test-1ml of sample solution was addad to 2ml of Mayer’s reagent.Formation of a dull white precipiotate indicates the presence of alkaloids. d.Hager’s test-1ml of sample was added to 3ml of Hager’s reagent.The formation of yellow precipitate indicates the presence of alkaloids.

2.Detection of Carbohydrates

a.Molish test-1ml of extract solution was added to 1ml of alpha-naphthol solution and concentrated sulfuric acid was added through the sides of the test tube.Purple or reddish violet colour at the junction of two liquids shows the presence of carbohydrates. b.Fehling’s test-1ml of extract solution was added to equal quantities of Fehling’s solution A and B.Upon heating the formation of a brick red precipitate indicates the presence of carbohydrates. c.Benedict’s test-1ml of sample solution was added to 5ml Benedict’s reagent and boiled for 2 minutes and cooled.Formation of red precipitate indicates the presence of carbohydrates.

3.Detection of Glycosides

a.Legal’s test-To the sample solution added 1ml pyridine and 1ml sodium nitroprusside and made alkaline.The formation of pink to red colour shows the presence of cardiac glycosides. b.Borntrager’s test-1ml sample solution was added to a few ml of dilute sulfuric acid.Boiled,filtered and extracted with chloroform.The chloroform layer was treated with 1ml ammonia solution.The formation of pink colour shows the presence of anthraquinone glycosides. c.Modified Borntrager’s test-to 5ml of extract added 5ml 5% ferric chloride and 5ml dilute hydrochloric acid.Heated for 5 minutes in a boiling water bath.cooled and added chloroform,shake well.Seperated the organic layer and added...
