ABSTRACT
The determination of etradiol levels to assess the menopausal status, ovarian function and fertility in females is essential. The challenge however, arises due to low concentrations of estradiol found in physiological samples from menopausal females.   This has lead to a need for the development of new methods with increased sensitivity. This project involves the development of a HPLC method for the detection of derivtaize estradiol. The detection limit of non-derivatize estradiol was determine to be 0.4382µg/ml. Derivatization of estradiol with dansyl cholride was unsuccessful due to degradation or incomplete derivatization and more work is needed to   optimise   this procedure.

INTRODUCTION
                      Determination of steroids in tissues or in biological fluids is necessary to elucidate the nature of many endocrine disease processes and thus and in the diagnosis and treatment. Steroids have strong biological activity at very low concentration (Picomolar and Nanomolar) in target tissues1. Immunoassays (IA) and radioimmunoassay (RIA) techniques are commonly used for determination of steroids2. Lack of specificity, limited dynamic range, and matrix effects are main drawbacks of IAs3, 4.   Estradiol is responsible for maintenance and the development of secondary gender characteristics and reproductive function in females. If the concentration of estradiol is low in women’s it can lead to disturbed puberty, oligoamenorrhea, and menopause6. For accurate diagnosis of disorders related to defective steriodogensis and various other clinical situation steroid profiling is very useful5.
• In the following, a literature review on the function, detection and measurement of one estrogens namely estradiol will be conducted.
