A. What is fermentation?
It is a metabolic process that allows the production of ATP without need for oxygen

B. Why is it important not to incubate the fermentation tubes beyond 24 hours?
You may not get accurate results. Although sugar is the primary nutrient used, when a microbe runs out of it, protein or other nutrients will be attacked This causes changes in the color of the medium because of the pH indicator added to detect acid production. When proteins are used, alkaline by-products are produced and the medium can changes colors. If you let these sugar tests go for more than a day, you risk the possibility that the color will have changed and you may misread the test result.

C. Why is phenol red added to the fermentation tubes?
Phenol red is added as a pH indicator. It will turn yellow below pH 6.8 and a darker-pinkish red above pH 7.4

D. Why do bacteria have differences in the carbohydrates they can ferment?
The ability of inability of a particular species to ferment a particular carbohydrate depends on the presence of the enzymes needed for a particular fermentation pathway. As it is in the DNA of an organism that codes for which particular enzymes that organism contains, the presence or lack of a particular enzyme is decided at the genetic level and varies by species.

E. Why does the formation of yellow color indicate fermentation?
Acid production produces a color change from red to yellow, indicating the organism is capable of metabolizing the sugar in the tube

F. What information can be gained by running a fermentation series on a particular microbe?
For each carbohydrate tested, the following information can be learned:
-can the organism ferment the particular carbohydrate
-if so, does it produce acidic end or by-products
-if so, does it produce gaseous products

G. What does a dark pink or red color indicate?

    The streak for isolation was chosen to see the formation of the different colonies and to see if the bacteria would cause growth on the plate. After observing the plate, it could be seen that the unknown had growth and formation of colonies. The Gram stain was chosen next to determine if the unknown was gram-negative or gram-positive. After looking at the gram stain under the microscope with the oil immersion it could be determined that the unknown was gram-negative due to the fact that the bacteria was pink in color and rod shaped. After determining the unknown was gram-negative, other bacteria’s were able to be eliminated from the options. The Triple Sugar Iron Test was chosen next because the unknown could be tested for glucose fermentation, lactose fermentation, sucrose fermentation and sulfur reduction. If the organism is able to ferment glucose and lactose and/or sucrose it will turn the medium yellow throughout (Leboffe & Burton 329). When the Triple Sugar Iron Test was complete, it showed a yellow butt, yellow slant and gas production. This determined that the unknown ferments glucose and one or both of the other sugars. The gas was produced by fermentation. The color change in the agar brought the unknown down to two options of unknowns, Escherichia coli or Hafnia alvei.     The two types of fermentation that are well known are alcoholic fermentation as well as lactic acid fermentation. Fermentation is vital for many organisms, such as yeast and bacteria, because it allows them to obtain energy required to carry on life processes.     In addition the indicators that were the most accurate were the pH paper and the phenalthalein. The phenalthalien was one of the most accurate indicators because the bright magenta pink color that a base substance turned was very prominent compared to the clear color that formed when the substance was an acid. The pH paper was the most accurate indicator because there was a color chart that showed specifically whether or not the substance was a base or an acid. The pH paper also got detailed enough to show whether or not the substance was pure/neutral or a strong base or acid. Furthermore, it was easy to tell that acids were 1-6 on the pH scale, water or neutral substances were 7, and bases were from 8- 14. The other indicators were not as accurate. The indicator phenol red always turned the substance either a yellow orange color or a red orange color, and that was sometimes a little difficult to figure out whether or not a substance was a base or an acid. The bronthymol blue did turn a yellow color in the presence of an acid, but sometimes the substance either turned a greenish color or had specks of blue in the substance.
