Biochemical Test Eneterobacteria
I. Purpose
The purpose of this lab was to become more familiar with Enterobacteriaceae Biochemical Tests.

III. Methods
Phenol Red Dextrose, Lactose, and Sucrose with Durham tube:
The procedure for inoculation and interpreted results were used from Chess + Handout. A table was made and observations of growth, gas, and color for each bacterium were placed in the table. The broths were incubated at 35ºC for 24 hours.
Note: three broths of each type of sugar were used.
Casein Plate:
The procedure for inoculation and interpreted results were used from Chess + Handout. A table was made and observations of growth and Zone of hydrolysis for each bacterium were placed in the table. The plates were incubated at 35ºC for 24 hours
Note: Only 1 plate of Casein was used which was divide like a pie into three sections. Starch Plate:
The procedure for inoculation and interpreted results were used from Chess + Handout. . A table was made and observations of growth and iodine test results from each bacterium were placed in the table. The plates were incubated at 35ºC for 24 hours

Litmus Milk:
The procedure for inoculation and interpreted results were used from Chess + Handout. . Two tables were made which recorded bacteria’s growth, color, gas, and consistency. The broths were incubated at 35ºC for 24 hours.
Note:
Gelatin Deep:
The procedure for inoculation and interpreted results were used from Chess + Handout. A table was made and observations of growth and liquid for each bacterium were placed in the table. The deeps were incubated at 35ºC for 24 hours.

Nitrate Broth with Durham Tube:
The procedure for inoculation and interpreted results were used from Chess + Handout. A table was made and observations of growth, gas, and degree of reduction for each bacterium were placed in the table. The broths were incubated at 35ºC for 24 hours Note:
TSA Catalase Slants:
The procedure for inoculation and interpreted results were used from Chess + Handout. . A table was made and observations of growth and H2O2 presence for each bacterium were placed on the table. The slants were incubated at 35ºC for 24 hours.
Note:
Enterotube:
The procedure for inoculation and interpreted results were used from Chess + Handout. . The results were recorded on a pre-made Enterotube identification handout. The Enterotube was incubated at 35ºC for 24 hours.
Note:
API
The procedure for inoculation and interpreted results were used from Chess + Handout. The results were recorded on a pre-made API identification handout. The API strip was incubated at 35ºC for 24 hours.
Note:
Once the index value was found, the number was used to search for the unknown in the API Quick Index Booklet. . An oxidation test was performed to make sure the unknown would perform positive tests. IV. Results
